Fluorine is an essential trace element for human health and life activities,which is harm to human health with excessive or little intake.Tea tree is a rich fluorine plant with rich fluorine in leaf,and previous research has found a new tea fluorine morphology,tea polysaccharide fluorine.Tea polysaccharide is an important active ingredient in tea,and the research of the combination of the tea polysaccharide fluorine can provide certain theoretical basis for tea polysaccharide's more reasonable,scientific and safe development and utilization.The research focuses on the assay method of total fluoride in tea and tea polysaccharide,influential factors of tea polysaccharide fluorine content,the morphology of tea polysaccharide fluoride,the main component content change of tea polysaccharide fluoride in purification,analyzing the effect of polysaccharide structure change for fluorine combined with polysaccharide in purification,the effect of metal ion for fluorine combined with polysaccharide.The main research results are as follows:1.The research of the assay method of total fluoride in tea and tea polysaccharideThe measured value of alkali fusion ashing method is higher than boiling water extraction and acid extraction method,which suggests that boiling water extraction and acid extraction can measure partial fluoride in tea and tea polysaccharide and can't reflect the actual total fluorine content.The fluorine recovery of alkali fusion ashing method is the highest at 83.04-94.40%,and RSD with alkali fusion ashing method is less than 3%.This result shows that alkali fusion ashing method has good recovery and high precision in measuring fluorine content,which is more suitable for measuring total fluoride in tea and polysaccharide.2.The research of influential factors of tea polysaccharide fluorine contentTaking tea leaf in the same environment as research object,the research of tea polysaccharide fluorine content effect by tea tree variety and maturity can eliminate the difference in fluorine content due to different planting environment.The result shows that tea polysaccharide content increases significantly with the increase of maturity in the same tea variety;There is also a significant difference on tea polysaccharide between different types in the same maturity.The experiment also researched the effect on tea polysaccharide fluorine content with different processing technology of tea,taking Wu Niu Zao no lignification red stem as materials in the same planting environment.Result shows that processing technology has significant impact on tea polysaccharide fluorine content with the same materials.3.The research of fluoride morphology in tea polysaccharideThe analysis of fluoride content and morphology in tea polysaccharide shows thatdifferent fluoride morphology in tea polysaccharide from high to low are water-soluble morphology at 81.3%-86.5%,fe-mn combined morphology at 81.3%-86.5%,interchangeable morphology,organic binding morphology and residue morphology at0.1%-0.2%.Wu Niu Zao and Fu Yun 6 can output much more tea polysaccharide fluorine,which is 86.8% and 91.2%,and it is the basis for fluorine safety evaluation of tea polysaccharide utilization.There is a significant positive correlation between total fluorine and different morphology fluoride content in tea polysaccharide,which shows that different fluorine morphology in tea polysaccharide can be interconverted under certain condition.4.The research of combination mode between fluorine and polysaccharideAs dialysis time increasing,the fluoride content in tea polysaccharide decreases.When the dialysis time reaches a certain level,the fluorine content in polysaccharide is basically stable.This shows that some tea polysaccharide fluorines are closely associated with polysaccharide.The polysaccharide fluorine content is reduced by a certain degree in the process of deprotein,dialysis and decoloration,which suggests that polysaccharide purification process has some effect on fluorine combined to polysaccharide.Neutral sugar,glucuronic acid,protein and fluorine are negatively correlated or unrelated,indicating that they have little efftct on fluorine combined to polysaccharide.However,aluminum,manganese,calcium and magnesium ions are positively correlated with fluorine,which indicates that they have great effect on fluorine combined to polysaccharide.The research of metal ion complexation for fluorine combined to polysaccharide shows that fluoride content increases significantly when tea polysaccharide directly complexing to fluorine,which suggests that tea polysaccharide has good ability of complexing to fluorine.The tea polysaccharide is combined with fluorine to form 5 types after combined with calcium,manganese,magnesium,aluminum and iron,and their fluoride content have a significant downward trend comparing with the polysaccharide directly combined to fluorine.The result suggests that metal ions may compete with binding site between fluorine and polysaccharide.In research of the effect of structural change on fluorine combined to polysaccharide in polysaccharide structure and its purification process,the infrared result shows that hydroxyl and amino group significantly offset in the process of polysaccharide purification,thus affecting polysaccharide fluoride content.As the process of polysaccharide purification increasing,polysaccharide gradually turns into a tight,smooth,flaky shape from a loose,fish-scale shape by analysis of scanning electron microscope onpurified polysaccharide,and this change may be related to fluoride reduction of polysaccharide in purification process.The thermal property analysis of polysaccharide sample in purification process shows that the substance aggregate state in polysaccharide may have changed,and the effect of this change on fluorine content in polysaccharide is still needed to research.Moreover,according to the result of high performance liquid chromatography,the proportion of monosaccharides in polysaccharide sample has changed as purification.And the effect of this change on fluorine content in polysaccharide is still needed to research.
