Inulin can inhibit postprandial blood glucose level,decrease oxidative stress and promote glucose decomposition.Ganoderma Lucidum Polysaccharides(GLPs)can improve insulin resistance,regulate the activity of glucose metabolic enzymes,increase insulin content,and reduce blood glucose level.However,it is not clear if the inulin and GLPs mixture had better antidiabetic effects than either inulin or GLPs alone.The aim of present study is to explore the hypoglycemia effect and molecular mechanism of the combination of inulin and GLPs.By exploring the inhibition of inulin and GLPs on the activity of a-glucosidase,the improvement of insulin resistance in HepG2 cells and the hypoglycemic effect on type 2 diabetic rats,the following results were obtained:(1)Inulin and GLPs can inhibit the activity of a-glucosidase,and the combination had a better synergistic effect.Compared with inulin or GLPs alone,the inhibition effect of a-glucosidase was significantly stronger than that of inulin and GLPs alone.The results showed that inulin combined with GLPs could effectively inhibit the activity of ?-glucosidase and reduce the postprandial blood glucose level.(2)Inulin and GLPs can certain effectively alleviate insulin resistance in HepG2 cell,and the combined effect is better than the inulin alone.When inulin with different concentrations was combined with GLPs,the glucose consumption,glycogen content,SOD and GSH-Px activities of the cells were all increased,and there were significant differences among the combination groups and the single inulin group(p<0.01).This indicated that inulin combined with GLPs could effectively alleviate insulin resistance in HepG2 cells.(3)The inulin and GLPs can effectively improve the glucose and lipid metabolism disorder in type 2 diabetes mellitus(T2DM)rats,and the combination effect of inulin and GLPs is better than that of inulin alone.After intervention for 5 weeks,the blood glucose and blood lipids index of combination groups significantly improved,the damage of liver and kidney obviously alleviated,and the capability of antioxidant markedly enhanced.In a word,inulin compound GLPs can improve the glucolipid metabolic disorders,repair the liver and kidney damage,reduce oxidative stress of the T2DM rats to some extent.(4)Inulin combined with GLPs can regulate the PI3K/Akt pathway in T2DM rats,and the effect of conbinations was better than that of inulin alone.Compared with the single inulin group,the combination group can significantly up-regulate the expression of PI3Kp85 and GLUT4 genes and proteins,and promote phosphorylation of Akt and GSK-3?,which indicated that the combination of inulin and GLPs can effectively promote glycogen synthesis and alleviate insulin resistance in T2DM rats by regulating the PI3K/Akt pathway.
