At present,with the advent of aging society,the development and utilization of anti-aging food has become a hot spot in China and even throughout the world.Aiming at the present situation,in this study,D-galactose induced aging mice were used as a model,and the anti aging effects of the active peptide-N were studied systematically and comprehensively by exploring their cognitive behavior,oxidative damage and neuroprotection,to provide theoretical basis for the research and development of new anti-aging food in China.The main conclusions were as following:1.Effect of active peptide-N on cognitive behavior of D-galactose induced aging mice's.The appearance of each group of mice was observed,activity peptide-N could significantly improve the mental state of D-galactose induced aging mice.Open field experiment,Morris water maze experiment and tight rope test were used to detect the effect of active peptide-N on the ability of locomotor activity,learning and memory and exercise coordination of aging mice.The results of open field experiment showed that active peptide-N could significantly increase the number of across the squares and the number of hind limb stand by 42.30%and 83.85%in aging mice(P<0.05 or P<0.01),it had the effect of improving the ability of motor exploration.In the Morris water maze experiment,the latency was significantly shortened by 65.09%,and the number of crossing the platform and the time spent in target quadrant obviously increased by 53.52%and 95.30%in the aged mice after intragastric administration of the active peptide-N(P<0.05 or P<0.01),it is found that the active peptide-N could improve the learning and memory impairment caused by aging.In the tight rope experiment,the active peptide-N could increase the maintenance time and conversion score of the aged mice on the rope by 77.72%and 74.10%(P<0.05 or P<0.01),and it had the effect of enhancing the coordination ability of the aged mice.2.Effect of active peptide-N on oxidative damage of D-galactose induced aging mice.The body weights of mice ware observed for nine weeks,the results showed that active peptide-N had no adverse effect on body weight.The organ coefficient of each group mice were measured,it was found that the cardiac coefficient and the brain tissue coefficient of the high dose groups were separately significantly increased by 18.26%and 11.90%(P<0.05),it indicated that the active peptide-N could reduce the atrophy degree of the heart and brain tissue of the aged mice.The main antioxidant enzyme activities and MDA content in serum and brain tissue were detected,the results showed that the active peptide-N could significantly increase the activity of SOD,GSH-Px and CAT by 15.77%,63.48%and 50.69%,and it decreased the content of MDA by 26.19%in serum(P<0.05 or P<0.01).The active peptide-N could significantly increase the activity of SOD,GSH-Px,CAT and T-AOC by 26.89%,30.45%,58.00%and 69.72%,and decrease the content of MDA by 26.72%in brain tissue(P<0.05 or P<0.01).It indicated that the active peptide-N could enhance the antioxidant capacity of D-galactose induced aging mice and alleviate the oxidative damage caused by D-galactose.3.Neuroprotective effect of active peptide-N on brain tissue of D-galactose induced aging mice.The biochemical markers such as MAO,AChE,NO,NOS in brain tissue were detected,the results showed that the activity of MAO and AChE in the brain tissue of aging mice induced by D-galactose were significantly inhibited by active peptide-N,the activity decreased by 23.20%and 25.81%(P<0.05).And the NO content and NOS activity in the brain tissue were decreased by 15%and 23.13%(P<0.05).It showed that the active peptide-N could alleviate the neurotoxicity in the brain of the model mice.Furthermore,morphological observation of the cerebral cortex and hippocampus of the brain tissue was carried out.It was found that the number of neuronal cell layers in the cerebral cortex and hippocampal of the mice treated with the active peptide-N increased and the levels of the cells were closely arranged,and the number of neurons was also increased,it indicated that the active peptides had a good neuroprotective effect on brain tissue of D-galactose-induced aging mice.In this study,which based on the analysis of the above behavioral experiments,biochemical indexs and histomorphology,the active peptide-N can improve the ability of autonomous activity,learning and memory ability and exercise coordination in D-galactose induced aging mice.It can also decrease the oxidative damage of the body and increase its antioxidant capacity.And it can reduce the neurotoxicity of the brain in aging mice.The active peptide has good anti-aging effect in D-galactose induced aging mice.It has a very significant potential development value in the anti-aging food.
