Cefluroxime is a broad-spectrum,semisynthetic cephalosporins.By blocking the synthesis of cell wall,it has inhibiting effected for a wide range of gram-positive and gram-negative bacteria and keeps stable to the majority of ?-lactamase.In industrial production,DAAC was synthesized from 7-AC A.Frist,side chain was introduced to 7-amino acylation and a 3-to carbamoyl-cefuroxime acid(DCCFA)was gotten.Then cefliroxime acid was obtained from three hydroxymethyl transformation,further cefuroxime sodium or acetate can be synthesized.Therefore,the production of DCCFA is a key step in the entire process..In the current production,there are some difficulties in the crystallization process of DCCFA.The progress of crystallization is too long,the grains too small and filter link too slow.In this work,the crystallization process of DCCFA were investigated systematically to improve the production efficiency and reduce the cost.The spherical crystal was dealt with the X-ray powder diffraction,And collection the crystal structure data.The cell parameters and crystal system were ensured by TREOR90 of Materials Studio.The DCCFA crystal structure was analysis by Powder Slove,The theoretical crystal habit of DCCFA was predicted by BFDH and AE.Crystallization process is a complex multiphase mass and heat transfer process,the driving force of the main crystallization process from the crystallization system in thermodynamic non-equilibrium.Therefore,The crystallization thermodynamics behavior is very important.In this work,Using the the laser monitoring observation technique to measure the solubility of DCCFA in water solvent and pH=2 and pH=4 and methanol+water,the experiment temperature from 278.15K to 318.15K under atmospheric pressure.Finally establish the solubility model and calculate the heat of solution in different solvent systems.Using the thermodynamic data to study the crystallization process of DCCFA.Crystallization kinetics of directly influence the size and distribution,In this work,The method of intermittent dynamic was used to study the crystallization kinetics,Then regression the nucleation rate and growth rate equation and discusses the influence of various factors.Based on the research of DCCFA crystallization thermodynamics and dynamics,Adjustment the amount of seed,crystallization temperature,stirring intensity,pH and crystal growing time ect,In order to improve the particle size and distribution.Ultimately,through experiment can gain the optimal process conditions to products the spherical DCCFA and meet the production requirements.
