Nepeta angustifolia C.Y.Wu is one of the commonly used drugs in the Tibetan medicine,mainly used in clinical spasm Jue coma,stroke,epilepsy,stroke,embolism sore hurt pain treatment,with good results.At present,there is very few research on Nepeta angustifolia C.Y.Wu in modern Chinese medicine.The volatile oil extraction process and quality standards for Nepeta angustifolia C.Y.Wu,conducted pharmacological research,and determined the molding technological conditions of Nepeta angustifolia C.Y.Wu dripping pills were studied in this paper.This paper studied the volatile oil extraction process and quality standards for Nepeta angustifolia C.Y.Wu,conducted pharmacological research,and determined the molding technological conditions of Nepeta angustifolia C.Y.Wu dripping pills.Different methods were used for the extraction of volatile oil from Nepeta angustifolia C.Y.Wu,including steam distillation,solvent extraction,supercritical fluid extraction,ultrasonic extraction and enzymatic extraction.Comparison of different methods was obtained by recovery rate.Orthogonal tests were used to optimize the extraction process for volatile oil of Nepeta angustifolia C.Y.Wu by steam distillation extraction(SDE)and supercritical fluid extraction(SFE).The chemical compositions of volatile oils obtained by SDE and SFE were identified by gas chromatography-mass spectrometry analysis.The optimization of the SFE were:extraction temperature,50 ?;dynamic extraction time,80min;pressure.28MPa;and modifier volume,200mL.The extraction rate was 2.32%.And the best conditions of the SDE were:crushing degrees(mesh).40;Extracting time,5h;soaking time,2h;water added,1500mL.The volatile oil yield was 2.32%.The oil obtained by SFE and SDE were analyzed by GC-MS,and the results showed small differences in the main components,but SFE extracts were found to be markedly advantages over steam distillation extraction at production rate.The establishment of quality standard of Nepeta angustifolia C.Y.Wu includes identification,purity and assay of pharmaceuticals.The microscopic identification and TLC method were adopted for qualitative identification.The results showed that with a petroleum ether and acetone(13:2)as outspread preparation,the TLC identification was distinct and the spots were clear.Moisture content,total ashes test,acid insoluble ash and ethanol-soluble extract were deremined to assess the quality of Nepeta angustifolia C.Y.Wu.The moisture content was not exceed 8%.the total ash content was lower than 14%,acid insoluble ash content was not exceed 5%and ethanol-soluble extractives not less than 5%.HPLC method for determination of pulegone in Nepeta angustifolia C.Y.Wu was presented.Results showed that there was a good linear relationship between the concentration of pulegone and area value in range of 0.008?0.4ug,and the correlation coefficients was 0.999 espectively.And pulegone content not less than 9.0%(mg/g).This method is rapid,simple and accurate.The SDE was used for determination of volatile oil,and results revealed that the volatile oil of Nepeta angustifolia C.Y.Wu must not be less than 0.50%(ml/g).Pharmacological studies proved Nepeta angustifolia C.Y.Wu volatile oil had anti-inflammatory,antipyretic and it could increase the intracellular oxygen concentration,which may be the mechenism of its cerebral protective effect.The cyclodextrin inclusion technique was employed to determine the optimum method of inclusion of volatile oil from Nepeta angustifolia C.Y.Wu.The optimum condition was:the proportion of volatile oil and 7-cyclodextrin was 1:6,inclusion temperature was 50 ?.and inelusion time was 4h.This method turns liquid state into solid state to reduce the volatilization of volatile oil.which is convenient and suitable for mass product.The best technical parameters of Nepeta angustifolia C.Y.Wu pills were investigated by orthogonal study.The optimized preparation conditions were as follows:with PEG4000 as optimal bases,optimal drugs and bases was 1:13,temperature of mixture was 80?,dropping speed was 45 d/min.0-5? dimethyl silicone oil as condensate,and dropping distance was 7cm.The preparation procedure is simple,feasible,stable and reproducible.
