The morbidity and mortality of Asian countries in breast cancer, prostate cancer and colon cancer and other malignant tumors are generally lower than the United States and other developed countries, this phenomenon is considered with Asians like to consume more soy foods. Many studies have found that soybeans, especially soy isoflavones, has significant cancer inhibition, but the mechanism has also been a research focus. In this study, we investigated the mechanism of soybean isoflavones in the epithelial-mesenchymal transition of colon cancer HT-29 cells.The MTT test indicated that soybean isoflavones was better effective on HT-29 cells,and presented the concentration dependence. The optimum inhibition concentration on HT-29 cells was 200?mol/L at the time of 48h. The results of DAPI staining, AO/EB staining indicated that soybean isoflavones inducing HT-29 cells apoptosis based on the typical apoptotic morphological changes. The results of DAPI staining, AO/EB staining indicated that SI inducing HT-29 cells apoptosis based on the typical apoptotic morphological changes, and the apoptotic rates significantly increased with prolonging the cultured time.By Transwell chamber invasion discovery, after genistein, HT-29 cells to reduce the number of invasive, well tumor necrosis factor-? effect increasing the number of cell invasion. RT-PCR results showed that genistein inhibits cell invasion and tumor necrosis factor-? promotes cell invasion. Cadherin immunostaining and E-cadherin immunofluorescence staining showed that genistein can inhibit the EMT by inhibiting the expression of E-cadherin and the expression of N-cadherin.The results of western blot showed that the effect of genistein on the expression of Notchl and the expression of PI3K in HT-29 cells decreased the expression of NF-?B p65 and decreased the expression of phosphorylated NF-?B p65, thus Bax/Bcl-2 were upregulated, and then Caspase8 cascade was induced to induce apoptosis of HT-29 cells.
