The soybean isoflavone is the second grade metabolic product in the process of the soybean growth.It has a range of biological activities,such as the preventing and fighting against cancer,resistance of biological oxidation,treatment of osteoporosis and menopause syndrome.Use soybean meal as the raw materials,with ethanol to extract the soybean isoflavones from the meal,neither effect the soybean meal feednutrition value,or can get the high added value of soybean isoflavones products,improve the comprehensive economic benefit of soybean meal.The paper about using soybean meal as the raw material,research and prepare 10%\40%and other higher purity soybean isoflavones production process.The extraction of soybean isoflavones adopt the normal temperature of soaks extraction,by single factor and orthogonal test,optimize the four factors of extracting soybean isoflavones from the soybean meal.:ethanol concentration,material to liquid ratio,temperature,and time,and get the best extraction conditions:extraction temperature 60?,extracting time 8 h,ethanol concentration 70%,material liquid than 1:8 g/mL,extraction rate 0.548%.10%soybean isoflavone products are preparated with using polyether sulphone organic membrane ultrafiltration soybean meal extract.Ultrafiltrate to emove the protein,polysaccharide,gelatin and other large molecules material,From the four aspects to optimize the ultrafiltration technology:the ultrafiltration membrane aperture,ultrafiltration pressure,temperature,material liquid flow.Then get the purity of 12.6%soybean isoflavone products.The best ultrafiltration technology is:ultrafiltration membrane aperture 5k D,ultrafiltration pressure 5 bar,ultrafiltration temperature 25?,material liquid flow rate is 11.8 mg/mL,the yield is 86.7%.40%soybean isoflavone products are preparated by the method of combining the polyether sulphone organic membrane ultrafiltration with macroporous resin purification.Before macroporous resin column purifying soybean isoflavone,first use organic membrane ultrifiltration extraction to remove the large molecules such as protein material.Through the single factor experiment,from D101,ADS-7,DM130 nine kinds of resin to get an optimization of the D101 macroporous resin as adsorbent,from the static and dynamic two aspects to investigate D101 exa mines macroporous resin adsorption influence factors,get the purification and optimization process,and ultimately get the purity of 41.21%soybean isoflavone products.The best process route as:temperature 25?,velocity is 1 mL/min,soybean isoflavone concentration is 1 mg/mL under the conditions of the column on adsorption.After the soybean isoflavones getting on,first use 1 BV deionized water to column on,then use 1 BV 10%ethanol with one of the impurities preli minary parts,and with 80%ethanol to continue elution and collect 20 to 60 mL part of the eluent,of soybean isoflavones were 65.7%.High purity soybean isoflavone products are preparated with the method of gradient elution.In the process of purifying soybean isoflavones with the column chromatography,use different concentrations of ethanol gradient elution preparation soybean isoflavone products,get the purity of the products of soybean isoflavones reached 67.65%.The gradient ellution process route is:first use 10%ethanol to wash,and then separately in 20%,30%,40%,50%ethanol to wash,collecting the 30%,50%ethanol part,get the product is of 67.65%purity,the products yield is 64.6%.
