Protein-polymer conjugates are the combination of proteins and polymers via specific sites or manners.When a protein is combined with a functional polymer which shows the biological activities,the resulting protein polymer-conjugates have the dual characteristics of polymers and proteins.The polymers and proteins both complement each other,which greatly broaden their range of applications.The bioconjugates based on protein/peptide grafted covalently into polymers have been shown a significantly effect to improve their applications and properties such as solubility,stability and biocompatibility.These macromolecules could be applied in the field of biomedicine and could show potential in many other areas,such as materials science and nanotechnology etc.It is highly popular novel material,which possesses the potentials of development in new polymer materials.It belongs to the interdisciplinary.This paper is divided into two parts.The first part is the synthesis of(2-(aminooxy)ethyl)2-bromo-2-methylpropanoate(ABM)which has an aldehyde and an amidogen group in two terminals respectively.Then,the N-terminus of BSA is transformed to an aldehyde through a biomimetic transamination reaction by 5'-pyridoxal phosphate(PLP).ABM is attached to the N-terminus of BSA,through a reaction between the aldehyde and the hydroxylamine to form a macroinitiator(BSA-Br).In the second part,poly(OEGMA)is directly grown from the protein macroinitiator by atom transfer radical polymerization(ATRP)in PB buffer.FT-IR?1H-NMR?UV-Vis and MALDI-TOF-MS measurements indicated that the compound and copolymers both were well controlled.The results showed that after modification by PLP the site specificity of the modification at the N-terminus of BSA was implementated and N-terminal peptide fragment with a bromine was observed for BSA-Br.Then in situ ATRP of OEGMA was successfully carried out from Mb-Br in PB buffer.At last,we got a site-specific(N-terminal)and stoichiometric conjugate(1:2)notably.
