Nanostructured drugs have become a hot topic in international research because of their unique advantages,which are different from traditional drugs.The preparation technology is an international cutting-edge high technology,which has great application value and theoretical significance.Subcritical water as a new type of green solvent,in many areas has a broad application prospects.In this paper,by studying the phase equilibrium properties of ciprofloxacin in subcritical water at different temperatures and pressures,the solubility parameters were determined and the appropriate solubility model was used to test the theoretical basis for predicting the dissolution characteristics of subcritical water.Based on the data of its dissolution characteristics,nanostructured ciprofloxacin granules were prepared by subcritical water combined with rapid precipitation technique,and a new green and environment-friendly nanostructured drug preparation technique was obtained.For the solubility of ciprofloxacin in subcritical water first of all,explored the equilibrium time of this factor.The total range of 100 ? to 170 ?,the temperatureset by the corresponding value of the drug to achieve the dissolution balance of the length of time required is different.The temperature goes high and the drug dissolves completely for a longer period of time.Take a look at the result,we selected 20 minutes as the required time.Then explored the temperature factor and found that the temperature becomes larger can cause ciprofloxacin to dissolve more in subcritical water.Finally,explored two factors,pressure and the addition of ethanol as co-solvent,the pressure has no impact on the solubility of ciprofloxacin,can not be called the factors of ciprofloxacin solubility in subcritical water.The addition of co-solvents enhances the ability of subcritical water to dissolve ciprofloxacin.For the preparation of ciprofloxacin nanoparticles by subcritical water,I explored the effects of many factors.Finally,the best conditions were as follows:temperature of subcritical water is 150 ?,temperature of deionized water is 0 ?,subcritical water and deionized water volume ratio is 1:3,concentration of co-solvent is 10%,concentration of surfactant polyethylene glycol concentration is 0.15%.The experimental results showed that the size of ciprofloxacin nanoparticles were the smallest and the particle size was about 50 nm.It is found that the subcritical water method does not change the chemical structure of the drug by comparing the infrared spectra of ciprofloxacin with the subcritical water treatment.XRD comparison of the two shows that the crystallinity of ciprofloxacin prepared by subcritical water method is reduced,which is amorphous and more easily absorbed by the human body.The dissolution of ciprofloxacin prepared by subcritical water method has been improved obviously by the in vitro dissolution of the two.
