In this study,Cyclocarya paliurus polysaccharide(CP)was isolated from leaves of Cyclocarya paliurus,and CP was sulfated modification to obtain sulfated Cyclocarya paliurus polysaccharide,named S-CP1-4.The basic physicochemical properties such as neutral sugar content,uronic acid content and protein content,molecular weight distribution,monosaccharide composition,FT-IR,UV spectrum and SEM images of CP and S-CP1-4 were conducted.The antioxidant activity in vitro was evaluated based on DPPH free radical system.The oxidative damage model of mouse macrophage cell line RAW264.7 was induced by Hydrogen peroxide(H2O2)in 1 ?moL/L.Meanwhile,the effects of CP and S-CP1-4 on the LPS induced mice inflammation were also detected.Based on physical and chemical properties and activity evaluation,the main results were as follows:(1)Sulfated modification can change the basic physical and chemical properties of CP including monosaccharide composition and molecular weight.The characteristic absorption peaks of S-O and C-O-S groups were observed,which could confirm that the sulfated group has been successfully combined into CP.CP is a kind of polysaccharides containing 63.77±1.55% neutral sugar,11.03±0.17% uronic acid and 8.23±0.78% protein.The sulfated group was non-detected in CP.However,S-CP1-4 containd neutral sugar of 42.41±2.55%,uronic acid of 13.41±0.69% and protein of 1.95±0.09%.Substitution degree of S-CP1-4 was 0.42.The results of SEM indicated that sulfated modification could cause a significant change to morphology of polysaccharides.But the monosaccharides composition analysis showed that the integrality about the main chain was protected.(2)Sulfated modification can enhance antioxidant activity of CP.Compared with CP,S-CP1-4 had more noticeable DPPH radical scavenging activity.S-CP1-4 could improve the viability of RAW264.7 cells induced by H2O2,as well as inhibite the lipid oxidation by down-regulating the level of MDA.Meanwhile,treatment with S-CP1-4 also increased SOD activity.(3)Sulfated modification can enhance anti-inflammatory activity of CP.S-CP1-4 was found to inhibit nitric oxide production,phagocytic activity and the release of interleukin(IL)-6 and IL-1? in lipopolysaccharide-treated macrophage cells,RAW 264.7.S-CP1-4 significantly decreased the secretion of IL-6 and TNF-? and the thymus and spleen indexes,and increased the production of IL-10 in lipopolysaccharide-treated mice.S-CP1-4 could better protect the liver by inhibiting the activities of alanine aminotransferase and aspartate aminotransferase,and malondialdehyde level while increasing the superoxide dismutase activity and total anti-oxidative capacity.
