Cyclocaryapaliurus whose leaves were rich in the flavonoidsbelongs to Juglandaceae.For their anti-inflammatory and relieving cough effects, the flavonoids were usually used in the treatment ofthe acute and chronic respiratory tract diseases.The flavonoids were nontoxic and widely spread in the nature.They can notbe synthesized which can bemetabolized quickly in the body. Recently it has become the hot point of extracting the flavonoids. Therefore, it’s important to establish an efficient extraction method. On the base of the dried leaves of Cyclocaryapaliurus, thisthesis focuses on the following four aspects:Ultrasonic coupling with theethanol extrat the flavonoids of the Cyclocaryapaliurus leaves by orthogonal test method. The extracting solution of the last step was filtrated through the ultrafiltration membrane. The flavonoid in the dialysate of the last step was absorbed by the macroporous resin and eluted by the ethanol. In the end the anti-microbe of the flavonoids from the Cyclocaryapaliurus leaves was studied.The results were as follows.1.The enzyme and ultrasonic extraction optimums. The single-factor experiments and orthogonal test show that:in the fixed conditions(particle size200mesh material, ethanol concentration75%, dual-band (45KHz), ratio of material to liquid1g/40mL), the temperature of the largest, followed by time, the impact of ultrasonic power is relatively small. Therefore the optimum conditionof extracting flavonoids in leaves of Cyclocaryapaliurusare:75℃,45min,450W.2. Studied the effects of pH, temperature, illumination, metallic ion, carbohydrate material, the oxidant and the reducing agent to the extraction of Cyclocaryapaliurus (Batal.)Iljinskaja by spectrophotometry. The result indicated that it is stabile for this extraction at pH7.0, and the color is the most wonderful. The flavonoids are relatively stable at low temperature, and the absorbance decreases with a rise of temperature. And the flavonoids’color faded easily with the increase of illumination. Na、Li、Zn、C、Mg、Al、Sn ions in the metallic ion solution have certain destructive effect to flavonoid, and it’s most unstablest with Cu and Sn ions. Regarding the carbohydrate material, the starch is the largest influentfactor. The influence of oxidant and reducing agent, it’s unstable by reducing agent.Staphylococcusaureus, Bacillussubtilis, Penicillium and Saccharomyces cerevisiae were selected to examine the anti-microbial activities of the purified flavonoids from the Cyclocaryapaliurus. Finally the minimum concentration of the purified flavonoids from the Cyclocaryapaliurus on inhibiting the microbe was determined. The result showed that the extract flavonoids from the Cyclocaryapaliurus didn’t have an obvious effect on the growth of Penicillium, Saccharomyces cerevisiae, Staphylococcusaureus and Bacillussubtilis. The antimicrobial activity of extract flavonoids from the Cyclocaiyapaliurus should be discusssed further.3. The prescription of the beverage made of Cyclocaryapaliurus leaves was studied, and the stability was also tested. The results showed that the ideal prescriptionis70%extraction liquid of Cyclocaryapaliurus,6%sucrose,2.8%honey and0.2%citric acid. The color of the product was bright orange, and it had flat aroma of Cyclocaryapaliurus and moderate sweetness and acidity. The experiment shows that starch, high temperature, strong illumination, metal ion, specially, ferric ion had an obvious effect on the stability of the drink. Therefore, the Cyclocaryapaliurus drinks should be preserved inside the containers without iron, zinc, aluminum, copper and calcium under low temperature in dark places.This research provides a reference for industrialization and rationalization of the flavonoids extracting from the loquat leaves. Fujian is one of the major provinces producing the Cyclocaryapaliurus. Nowadays, most of Cyclocaryapaliurus leaves are treated as waste. Therefore, there will be good social and economic achievements if we use this natural resource effectively.
