Atrazine is a trazine pesticide, anilofos, an organophosphorus pesticide. Both of these are used to inhibit the growth of weeds in farmland. The inappropriate use of pesticides poses a series problem of pesticide residues, and hazardes the safety of environment and cereal, even for human health .This paper aims to study a rapid and efficient separation and detection method to detecting pesticide residues in the environment and cereals.The dendritic silver nanoparticles were used as the Raman enhancement substrates,and the melamine was as the Raman signal,labeled the hapten.Established a methord that competitive immune combined surface - enhanced Raman spectroscopy to detect pesticide residues in the environment and grains.The optimal scanning parameters of the Raman spectra were obtained by several experiments: the scanning time was 10s, the scanning times were 3 times, and the laser power was 0.5% -5%. The dilution factor of the antibody was 10 times dilution, the dilution factor of the labeled hapten was 100 times diluted, the incubation time was 25 minutes, and the number of elution of PBST buffer was 2 times.There are two kinds of target,atrazine and anilofos.The solution gradient concentratione from 0.01ng/mL to 100?g/mL was as the abscissa,the Raman intensity of melamine was as the ordinate to established standard curve. The calibration curve of atrazine was y=-7079.3x+26571, R2=0.9688. The standard curve equation of atrazine was y=-4871.6x+17999 and R2=0.9705. The detection limits for both methods were 0.01 ng /mL.The results of immunoassay combined with surface-enhanced Raman spectroscopy for the detection of pesticide residues in the environment and grain are short,sensitive, specific and with the high recovery rate for the actual sample spiked .The method is suitable for quantitative determination of many pesticide residues.
