Worldwide,iron deficiency anemia(IDA)is a common disease which is due to lack of iron elements and harmful to human health.Therefore,it is of great significance to study the synthesis and production of iron supplements.Ferrous sulfate,ferrous succinate and ferrous gluconate are common iron for clinical treatment.However,these iron desorb free iron ions in the gastrointestinal tract which have a stimulating effect on the gastric and intestinal mucosa.Nausea,vomiting,abdominal pain,diarrhea and other gastrointestinal reactions appear as well.Polysaccharide iron belongs to the third type iron supplement,and it can be effective in the treatment of iron deficiency anemia.It has the advantages of high stability,good water solubility,easy absorption,high bioavailability and low toxic side effect.Currently,the market has been listed on the polysaccharide iron complex in the United States listed Niferex and Hong Yuanda in China.In this paper,we prepared the polysaccharide iron complex with non-toxic and cheap raw materials by using the method of chemical synthesis.Response surface methodology(RSM)was used to investigate the complex reaction of six hydrated ferric chloride with syrup and various factors of alcohol precipitation and washing process.It is able to optimize the operation process to realize the mass production of iron polysaccharide complex.First of all,we prepared the polysaccharide iron complex by six hydrated ferric chloride and saccharide.By means of comparing the complex reaction of glucose and syrup with six hydrated ferric chloride,the latter was selected as complexing reagent.The effects of reaction temperature,pH value,time,feed ratio and stirring speed on the complexation reaction were investigated by single factor experiments.Three factors,including reaction temperature,pH value and reaction time,were determined at last.The optimal process for the synthesis of polysaccharide iron complex was determined by response surface methodology:the reaction temperature is 71.2?,the pH value of the system was about 10.98,and the reaction time was 3.48 h.The iron content of polysaccharide iron complex was 42.93%.Secondly,ethanol was used for the alcohol precipitation of polysaccharide iron complex.We used single factor experiments and response surface experiments to investigate the effects of the ratio of ethanol,centrifugal speed,centrifugal time and the amount of dissolved water on the alcohol precipitation and washing.The optimum process conditions of ethanol precipitation and washing of polysaccharide iron complex were optimized:absolute ethanol volume ratio was 69.7%,the optimal values of centrifugal speed and centrifugal time were 4960 rmp,10.6minand the yield was 39.90%.Finally,the structure of the product was analyzed by melting point determination,UV,IR and HPLC.
