Iron deficiency anemia is the most common of all anemia types and it is prevalent worldwide.Insufficient iron intake,reduced absorption and increased demand can lead to insufficient iron storage in the human body,which can not meet the needs of normal erythropoiesis and lead to iron deficiency anemia.According to the WHO,the incidence of iron deficiency anemia in adult women is higher than that in men,and the incidence of iron deficiency anemia in pregnant women is the highest.In addition,many children also suffer from iron deficiency anemia.Iron deficiency anemia patients can appear fatigue,anorexia,decreased immunity and other symptoms,which will seriously affect the health of the body.The use of iron supplement drugs is the most effective way to treat iron deficiency anemia.Therefore,the development of a high bioavailability,safety,effective and low-cost iron supplement drug is very valuable.At present,scientists have successfully developed three generation of iron supplement drugs.The first and second generation iron iron supplement drugs have good effect on iron supplementation,but they have great side effects on gastrointestinal tract and metal odour.The effect of the third generation of iron supplement drugs on iron supplementation is remarkable.Furthermore,they have less side effects,good stability,good taste and high bioavailability.Polysaccharide iron complex belongs to the third generation of iron-supplementation drugs.Polysaccharide iron complex is a new type of oral iron supplementation agent,which has a remarkable clinical effect in the treatment of iron deficiency anemia.Polysaccharide iron complex capsule Niferex has produced by Watts Pharmaceutical Factory and sold in our country.At present,a sort of polysaccharide iron complex capsule has been produced and sold in China.However,the reactor used is only1000 L,and the yield is only 36.7%.Therefore,it is of great significance to study the production process of polysaccharide iron complex,using larger reactor and further improving the yield.In this paper,non-toxic,cheap corn starch hydrolyzed syrup and six hydrated ferric chloride were used as starting materials in the synthetic process of corn starch polysaccharide iron in 5L reactor.The effects of reaction temperature,pH value of reaction system,reactiontime,stirring speed on iron content and stability of corn starch polysaccharide iron were studied by single factor experiment and response surface optimization method.And the optimum reaction conditions were: reaction temperature 75 C,reaction pH 11.00,reaction time3 h.Through the experimental study,the effects of the ratio of anhydrous ethanol,centrifugal speed,centrifugal time and the ratio of resoluble water on the yield of the product were analyzed,and the optimum conditions of separation and purification were determined.Then the process magnification was carried out,and the selection of production equipment and the key quality control point process were carried out.The effect of spray drying on the yield of corn starch polysaccharide iron was analyzed by experimental study.As a result,the production process route of corn starch polysaccharide iron in 3000 L reactor was determined.The iron content of the polysaccharide iron complex synthesized by the optimized technological route was 43.65 % and the yield was 41.48 %.Finally,the characteristics of corn starch polysaccharide iron were identified,including physical and chemical properties,structure confirmation and impurity analysis.
