Auricularia auricula polysaccharide possesses a large variety of biological fuctions.In this study,polysaccharides were extracted by ultrasonic-assisted hot water,and the bioactive polysaccharide fragments were screened by ethanol classification.This fragment was used for sulfation.The polysaccharides were used for sulfation to obtain sulfated Auricularia auricula polysaccharides.The structure and physiological activity of SNAAP were studied.Ultrasonic-assisted extraction of polysaccharides from Auricularia auricula,the optimum conditions were as follows: ultrasonic time 18 min,ultrasonic power400 W,ratio of material to liquid 1: 138,the Auricularia auricula polysaccharide yield?34.67 ± 0.32?%.Compared with the common water polysaccharide,the polysaccharide obtained by ultrasonic assisted extraction had higher antioxidant activity.NAAP40,NAAP60 and NAAP80 were obtained by ethanol classification.The antioxidant activity of NAAP80 was the best.The NAAP80 polysaccharide was modified by chlorosulfonic acid-N,N-dimethylformamide method,the optimum reaction conditions were as follows:reaction time 3 h,reaction temperature 51.06 ?,CAS-DMF reagent ratio 1:5.97,the substitution degree was 0.514.The IR spectra of SNAAP showed that the maternal characteristic peaks of polysaccharides were intact and there were two characteristic absorption peaks of sulfuric acid.SNAAP had strong absorption peaks at 1247.70 cm-1 and 816.76 cm-1,indicating that NAAP was successfully sulfated by CAS-DMF.HPLC analysis showed that the relative molecular weight distribution of SNAAP was uniform and the distribution was relatively narrow.The results of the monosaccharide composition showed that SNAAP was composed of mannose and glucose at a molar ratio of 1.7: 1.Scanning electron microscope analysis showed that the surface morphology of SNAAP was smooth and neat,with irregular fiber and filamentous deposits.The anti-radiation effect of sulfated Auricularia auricula polysaccharides was studied by establishing60Co-? radiation damage model in mice.After60Co-?radiation,SNAAP can improve radiation-induced oxidative damage.Radiation protection is mainly played in three ways: SNAAP improved the body weight ofmice,effectively improved the organ index of mice,increased monocyte phagocytosis,increased spleen lymphocyte proliferation,thereby improving the mouse immune system,effectively prevented radiation-induced oxidative damage.SNAAP could significantly increase the levels of SOD and CAT in serum,increased the activity of GSH and decreased the content of MDA in lipid peroxides.SNAAP group can effectively reduced the content of carbonyl protein in mice,decreased the levels of creatinine and urea nitrogen in mice.It played an effective radiation protection role.
