20(S)-Popopanaxatriol(PPT),a ginseng saponin,extracted from ginseng.American ginseng or Panax has good effect in regulating blood sugar,anti-aging and anti-fatigue.Especially in anti-tumor,inducing tumor cell differentiation or other aspects,the effects are more significant.Given poor solubility of PPT,the concentrations are often lower than required therapeutic purposes,which affects the utilization efficacy of the drugs.Cyclodextrin(CD)and its derivatives with unique structure,internal hydrophobic,outer hydrophilic,enable to embed insoluble drugs or groups into their molecular cavities in the way of covalent bonds,increasing the hydrophilic of the complexes and enhancing the drug's water solubility and bioavailability.In recent years,cyclodextrins have drawn significant attentions to the field of supramolecular chemistry for their improving the solubility of insoluble drugs.At the same time,cyclodextrin and its derivatives as a drug carrier for study on drug and BSA interactions,not only help to control the drug absorption,distribution and transportation process in vivo,to explore the incidence of certain diseases Mechanis m in molecular level,to find effective therapeutic drugs,but also they are of great significance in the field of protein science,genetics,pharmacology and pharmacokinetics and other research areas.In this dissertation,several CD derivatives with PPT supramolecular system were synthesized,which were further studied in the reaction with BSA.1.6 kinds of modified ?-CDs(host 1-6)were synthesized,and 4 kinds of supramolecular systems with PPT were prepared,by the characterization of 1HNMR,XRD and other methods.According to the 2D ROESY spectrum,the possible binding modes were speculated.By the way of UV titration,stability constant and inclus io n ratio of the host and the guest were obtained.The results show that the inclusion ratio of supramolecular systems are 1:1.2.The interaction between PPT complexes and BSA were investgated by fluorescence spectroscopy.The results showed the main forces of the binding process of PPT and BSA were hydrogen bond and vander Waals force,and the processes were static quenching.Simultaneous fluorescence spectroscopy and binding distance experiments indicated the supramolecular system acts on the tryptophan residues of BSA,and dipole-dipole nonradiative energy transfer could also cause fluoresce nce quenching of BSA.3.Through the study of the supernatant anion scavenging ability of the host1/PPT complex,it was found that the inclusion compound has the ability of anti-oxidat io n.When the concentration of the inclusion compound at 0.05-0.08mmmol/L,the supernatant anion scavenging increased rapidly.After 0.08mmol/L,it became steady and reached the top value about 52%.
