DNA is a molecular genetic instruction and it can guide the biological development and functioning.In order to explore new DNA probes and antitumor drugs,many research on the interaction of DNA and small molecules has been did in recent decades.Porphyrins have been reported a lot in the field of DNA probes,nucleic acid enzyme activity and photoinduced electron and energy transfer process simulation research in natural photosynthesis.However,studies on the nteraction of corrole complexes,especially non-metallic corrole complexes and DNA are rare,this need to be further explored.Based on the above research background,this thesis focused on DNA binding,photonuclease activity and cytotoxicity toward tumor cells of phosphorus corrole complexes.The main work as follows:Firstly,10-(4-hydroxylphenyl)-5,15-bis(pentafluorophenyl)corrolato(trans-dihydroxo)phosphorus(V)1-P,10-(3-hydroxylphenyl)-5,15-bis(pentafluorophenyl)corrolato(trans-dihydroxo)phosphorus(V)2-P and 10-(2-hydroxylphenyl)-5,15-bis(pentafluorophenyl)corrolato(trans-dihydroxo)phosphorus(V)3-P were synthesized and characterized by UV-Vis,NMR and MS spectra.Secondly,the interaction between 1-P and calf thymus DNA(CT-DNA)were studied by UV-Vis absorption spectroscopy,fluorescence spectroscopy,circular dichroism spectroscopy and viscosity experiments.Results revealed that 1-P bind to CT-DNA via an outside binding mode.Agarose gel electrophoresis experiments showed 1-P display good photonuclease activity.Inhibitor testing revealed that hydroxyl radical,singlet oxygen and superoxide anion were the active species in the DNA photocleavage.1-P stands for a new class of non-metal artificial nuclease.1-P is localized in cytoplasm and exhibits low dark cytotoxicity to H460 and A549 tumor cells,but exhibits high photocytotoxic activity against them.Thus,1-P may become the promising candidate for non-invasive treatment of cancer in PDT.Lastly,the interaction between 2-P and calf thymus DNA(CT-DNA)were studied by the same spectroscopic methods.Results also revealed that 2-P bind to CT-DNA via an outside binding mode.Agarose gel electrophoresis experiments showed 2-P display good photonuclease activity.Inhibitor testing revealed that hydroxyl radical,singlet oxygen and superoxide anion were the active species in the DNA photocleavage.
