Biomimetic chromatography,which uses the derivatives of bioactively molecules as the functional monomer to prepare a chromatographic materials?in the broadest senses?,it also can be refer to immobilized artificial chromatography?in the narrow senses?.As part of biominimetic chromatography,immobilized artificial chromatography have been widely used biofilm simulation,which have some applications in predition of drug-membrane perpeability,purification of proteins and separation mediums for small anlytes.Monolithic column,which be describes as “the fourth grneration of chromatographic column”,compared with traditional packed columns,with the merits of facile preparation procedure,high permeability,fast mass transfer values and easy modification,monolithic column have proved to be an effective alternative column technology of traditional packed columns.There are few of biomimetic monolithic column based on the derivatives of phospholipids,but their exsits some drawbacks in the phospolipds-based packed column.Based on the basic research foundation,so this paper intends to target design and prepare of different types of new organic polymeric phospholipids functionalized monolith.Chapter one,firstly,introduantion to the theoretical basis and applications of the biomimetic chromatography;then,the progress and current situation of the monolith column,including silica-based,polymer-based,hybrid-based monolith have been introduced,the hydrophilic interaction and mixed-mode polymer-based monolithic column made a detailed introduction.Finally,a detailed review to organic-based monolith prepared by a single crosslinker monomer.Chapter two,a single chain C12 phosphatidic aicd-based functional monomer?12-Methacryloyl dodecylphosphatidic acid,MDPA?have been semisynthesis based on the main components of biomembranes: phospahtidic acid.Poly?MDPA-co-EDMA?monolith was prepared of MDPA.Firstlly,the preparation condition were optimized by semi-empirical method,to obtain a satisfactory monolithic column.Then,the physical and chemical properities and chromatographic performances of monolithic column have been investigate,the results demonstrate that both cation-exchange and hydrophobic interactions could contribute significantly to the overall retention of charged analytes on this monolithic column.This could increase the separation power and degree of freedom in adjusting the separation selectivity compared to single cation-exchange or reversed-phase mode.Finnaly,some different class of standard analytes have been used for the inspection of the separation power of monolithic column.Chapter three,a bi-crosslinking C2 phosphatidylcholine-based functional monomer?2-?methacryloyloxy?ethyl-[N-?2-methacryloyloxy?ethyl] phosphorylcholine,MMPC?have been semisynthesis based on the main components of biomembranes: phosphatidylcholine.Poly MMPC monolith was prepared of single crosslinker functional monomer MMPC.Firstly,in order to elucidate the effect of the polymerization time on the morphology of nontraditional organic based monolithic support materials.Porosity parameters like poresize-distribution,total porosity and the specific surface area were investigated and then linked with the chromatographic analysis.The result suggested that monolith can be prepared within one hour.Then,the preparation condition were optimized by semi-empirical method,to obtain a satisfactory monolithic column.Then,the physical and chemical properities and chromatographic performances of monolithic column have been investigate,the results demonstrate that both weak electrostatic interaction and hydrophobic interactions could contribute significantly to the overall retention of charged analytes on this monolithic column,the results demonstrate that HILIC mechanism was determined over the ACN content range from 95-20%,which indicates the high degree of hydrophilicity.Finally,some different class of standard analytes have been used for the inspection of the separation power of monolithic column.Chapter four,the summary and outlook of thesis.
