In the environment too much steroid hormone of health problems take the public attention.Ctestosteroni bacteria secrete, 3?-HSD and 3,17?-HSD is decomposition of steroid response in the important function of two kinds of enzymes. By studying some regulatory factor for , 3?-HSD and 3,17?-HSD expression regulation, to provide more theoretical basis for the degradation of steroid hormones.This article will TetR gene by polymerase chain reaction from bacillus testosterone tufting order full sequence amplification. In order to prove the function of the gene, to TetR cloned into pET - 15b, pUC19, pK18 this several different expression vector and with,3?-HSD and 3,17?-HSD regulation sequence of plasmid transformation, with the analysis on steroid hormone induced the expression of TetR regulation function.By fluorescence detection and ELISA test, we found that TetR expression in the total transformation of bacteria, 3a-HSD protein expression than not compared to TetR gene of bacteria decreased obviously. Also in co-transformation TetR expression fungus in3,17?-HSD protein expression than not compared to TetR gene of bacteria decreased obviously. Show TetR is 3a-HSD and 3,17?-HSD suppression. And TetR to 3,17?-HSD respective area in the P2. And testosterone can remove TetR protein for 3a-HSD protein and 3,17?-HSD inhibition of protein expression. Further preparation of TetR polyclonal antibody by ELISA test TetR protein expression is not affected by the regulation of steroid hormones.
