In recent years,the development and production of new anti-tumor,anti-cancer,anti-oxidation,anti-aging and other new marine medical food has become a hot topic of concern.Detection of active ingredients in marine medical food always using high performance liquid chromatography,liquid chromatography-mass spectrometry and gas chromatography-mass spectrometry and so on.However,these methods usually require complex pretreatment: tedious operation,long time and sample contamination,so the technology can not meet the requirements of safety production and on-site rapid detection in supervision progress of marine medical food.The atmospheric pressure open-air mass spectrometry ion source can be used for rapid analysis of complex matrix samples without sample pretreatment.In this paper,a new droplet spray mass spectrometry ion source is developed based on cover slip as the substrate for sample loading,and the technology of new droplet spray ion source mass spectrometry was constructed.Aiming at different forms(solid,liquid)of marine medical food,a rapid quanlitative analysis method for active ingredients in marine medical food was developed based on new droplet spray ion source.A new droplet spray mass spectrometry ion source based on the cover slip tips as the substrate of the sample was developed,and a technology of new droplet spray ion source mass spectrometry was constructed.Optimizing the relevant parameters to obtain reliable,stable and efficient ionization efficiency.Based on this,further studied the ionization mechanism of droplet spray ion source.Taking cyclic adenosine monophosphate for injection,adenosine disodium triphosphate for injection and ademetionine1,4-butanedisulfonate for injection as research targets,the result tested the feasibility of rapid analysis active ingredients in marine medical food using the technology of new droplet spray ion source mass spectrometry.On the basis of the technology of new droplet spray ion source mass spectrometry,a rapid quanlitative analysis method for active ingredients in different forms(solid,liquid)of marine medical food was developed based on new droplet spray ion source.Taking taurine as the research target,the effects of ionization mode,spray solvent,positioning and spray voltage et al.on the response of taurine were investigated.The technologies of DSI-MS were constructed aiming at solid and liquid samples.Based on this,the taurine samples from six different manufacturers(three solid samples,three liquid samples)were analyzed qualitatively and the active ingredients in the six samples were quickly and accurately detected.Based on the technology of new droplet spray ion source mass spectrometry,a rapid quantitative analysis method for active ingredients in marine medical food was developed on the basis of new droplet spray ion source.Taking the vidarabine monophosphate as the model,which was the phosphorylation product of vidarabine extracted from spunge.The effects of ionization mode,positioning and spray voltage et al.on the response of vidarabine monophosphate were investigated.Taking characteristic fragment ion(m/z 211)of the vidarabine monophosphate as the quantitative ion by MS/MS,the linear regression equation was established: y = 96.44 x-389.08.The linear correlation coefficient R2 was 0.99,and the limit of detection(LOD)was 1.7 ng/mL.The recoveries ranged from 91.7% to 111.0% when the spiked concentration was 5-50 ng/m L.Based on this,the active components of the vidarabine monophosphate for injection of longfu brand,datong brand and xianqiang brand were quantitatively analyzed.The contents of active ingredients were 18.06,19.19 and 20.16 ng/m L,respectively,and the relative standard deviation(RSD)was 6.6%,6.6% and 6.9%,respectively.In this paper,a new technology of droplet spray ion source mass spectrometry was developed based on the cover slip tips as the sbstrate of the sample loading.Aiming at the needs for rapid analysis of active ingredients in marine medical food,a rapid quanlitative and quantitative analysis technology for active ingredients in marine medical food was developed based on new droplet spray ion source.And this technology has been successfully used in quick qualitative and quantitative analysis of the different forms of marine medical food.The technology has the advantages of strong resistance to matrix interference,short analysis time,no sample contamination,stability and simple operation.This paper provides the theoretical and practical basis for the rapid detection and analysis of the active ingredients in domestic marine medical food,which help to the safe production and standardized production of marine medical food and promote the establishment of relevant industry standards,and further promote the development and utilization of marine medical food.
