Objective: Illicium macranthum A.C.Smith belongs to the genus Illicium L.in the Illiciaceae plants,and Illicium species are the only genus of the Illiciaceae plants.The pericarps of Illicium species are rich in shikimic acid,which is relatively expensive in price and widely used in the area of bio-medicine.The Swiss pharmaceutical company Roche ever took it as basic raw materials of producing the antiviral drug Tamiflu.In this paper,in order to determine the methods for determination of the content of shikimic acid in the pericarps of Illicium macranthum and acquire the optimum technological condition for the reflux extraction of shikimic acid.Main research contents are as follows:Methods: 1.the methods for determination of the content of shikimic acid in the pericarps of Illicium macranthum An HPLC method for the determination of the content of shikimic acid is established in this research.This article examined chromatographic condition,system suitability,linear relationship,precision,repeatability,stability,average recovery,limits of detection(LODs)and limits of quantification(LOQs)for determination of the content of shikimic acid of Illicium macranthum to verify the methods for determination of the content of shikimic acid.2.The Central composite design-Response surface methodology is conducted to optimize the design of the extraction process of shikimic acid The reflux method is used to extract the shikimic acid and an HPLC method is established to determine the content of shikimic acid in this research,meanwhile,the trend line about the content changes of shikimic acid and factors-level table are acquired through single-factor experiments.The content of shikimic acid is taken as indexes,and Composite Design of response surface method was applied to optimize the reflux extraction process for shikimic acid of the pericarps of Illicium macranthum A.C.Smith,and the mathematical model between the factors and the response values was builded,then the feasibility of the model was further confirmed by the magnifying test and the validation test.Results: 1.the methods for determination of the content of shikimic acid in the pericarps of Illicium macranthum The linearity of shikimic acid is good,and the results of the experiment were conducted on precision,repeatability,stability,recovery of sample,test limits and quantitative limits are all in the relative error range,so the method of determination of shikimic acid was completed.2.The Central composite design-Response surface methodology is conducted to optimize the design of the extraction process of shikimic acid The best extraction condition of shikimic acid: soaking for 12 h by sealing up them on the optimal extraction conditions of methanol concentration,liquid-to-materia ratio were 25%,15:1,respectively,then on the optimal extraction conditions which the temperature was 70?,extracting 1 hours and one time;under these conditions,the extraction capacity for shikimic acid of the pericarps of Illicium macranthum was 92.5466mg/g,bias between observed and predicted values was less than 3%,and the correlation coefficient of the binomial fitting is 0.9702,so the model fits well.Conclusions: 1.High performance liquid chromatograph is highly efficient,selective and sensitive to the separation efficiency of shikimic acid,and the amount of shikimic acid measured is accurate.2.the technological design on optimizing the extraction process of shikimic acid which conducts a central composite design(CCD)and the response surface methodology(RSM)is simple to operate,and it is stable,meanwhile the extraction rate is higher and the prediction is good,it can be widely applied in the field of extraction of Chinese medicine.
