Objective : To investigate the damage of different concentrations of PM2.5 to AM(alveolar macrophages)of rats and the effect on the level of HO-1.Methods:PM2.5 was collected from Shijiazhuang city,Hebei province in haze weather and made it into different concentrations,the PBS buffer was used as negative control group.The AM which was extracted the from Wistar rats' bronchoalveolar lavage fluid(BALF),was exposed to different concentrations for 8 hours.The cell viability was detected using the MTT assay,and the level of HO-1 was detected by ELISA.Data analysis: using SPSS 21 statistical software for statistical analysis of all data.The data of normal and normal variance of measurement data were analyzed by single factor analysis of variance(One-Way ANOVA),and the comparison between the 5 groups was made by using the SNK method,and P<0.05 was statistically significant.Results:As the concentration of PM2.5 increased,the survival rate of AM was decreased gradually,but there was no significant difference between the50?g/mL group and the negative control group,but there was a significant difference in the high dose groups compared to each other(P<0.05).While the level of HO-1 was increased as the concentration of PM2.5 increased,these was a statistically difference in each group(P<0.05).Conclusion:The more serious haze was,the more damage to the AM.The level of HO-1 got higher with the concentration of PM2.5 increased,which showed it's protective effection under the stimulate of haze.
