Dark tea,one of the six major types of Chinese tea,which is belonging to the post-fermented tea and made by fermentation method.The mainly procedures are primary tea matching,stack screening,steaming and ottawa heaping,pressing setting,flooding and drying,Product Packing.According to the region can be divided into Hunan dark tea(Anhua dark tea),Sichuan Tibetan tea(Kang brick tea),Yunnan dark tea(Pu-erh tea),Guangxi Liu Pao tea,Hubei dark tea and Shanxi dark tea(Fuzhuan brick-tea).The functional ingredients of tea polyphenols,alkaloidTea,polysaccharide and L-theanine have anti-obesity and hypolipidemic,digestion,enhance capillaries,antioxidant and other functions.One of the most important features isEurotium cristatuminFuzhuan brick-tea.Because the production process was tedious,the fermentation period was long and the high costof extract functional ingredientsfrom Fuzhuan brick-tea driectly.Therefore,a solid-state fermentation technology was urgently needed to prepare the functional ingredients of dark tea.1.This part explorelarge-scale preparation of dark tea materials by solid-state fermentation technology,then the functional ingredientswere extracted from dark teaeffectively,in order to increase the content of active ingredients,the blue lightirradiation and solid-state fermentation interferencetechnology were added to solid-state fermentation process.Three different samples(FHZHQ-1,ZCFJ-1,LGJB-1)were extracted by ethanol extraction method.Ethyl acetate and n-butanol extracts were analysised by HPLC.The results showed that The peak area of EGCG from three samples was5020mAU*s,3715mAU*s,8286mAU*s.The peak area ofCAF was 8578mAU*s?6005mAU*s?10474mAU*s.The peak area ofEmodin was 2197mAU*s?872mAU*s?1667mAU*s,respectively.The total peak area of all extractive was 6.6349 mAU*S×104?6.3680 mAU*S×104?13.2257 mAU*S×104.There were four new peaks at 11.174 min,12.071 min,13.631 min and 16.906 min.Shows that the solid-state fermentation technologyof dark teaadded with blue light and distraction,not only to increase the content of EGCG,CAF and total fermentation product,but to producenew substances in the transformation process of dark tea with E.cristatum.2.We took samples of FHZHQ-1 and LGJB-1 as the materials for transcriptome sequencing,combined with the rice fermentation product(DMFJ-1)of the E.cristatum.Received 51630 transcriptsusing RNA-seq method on the Illumina HiSeqTM 2000 platform,and 19749 unigenes were annotated.Of these,9169 unigenes were annotated into metabolic process of the GO database,and 2465 unigenes were annotated intometabolism of the KEGG database.Analysis of gene differential expression three pairwise samples.The results showed that there were 1679 differential genes between DMFJ-1 and FHZHQ-1 samples,980 differential genes between DMFJ-1 and LGJB-1,2417 differential genes betweenFHZHQ-1 and LGJB-1,of which 1553 down-regulated genes and 864 up-regulated genes.Indicating that the difference between FHZHQ-1 and LGJB-1 was the most significant,and.Thus,the differential expressiongenes were analyzed with enrichment.Photosystem,photosynthesis and oxidation-reduction process were enrichment in the GO databasesignificantly.Photosynthesis and photosynthesis-antenna protein were enrichment in the KEGG databasesignificantly.So that,blue light irradiation and stirring stimulation added to the solid-state fermentation system of dark tea had a significant effect on the secondary metabolism of E.cristatum,and mainly focused on photosynthesis and enzyme activity.3.Functional ingredients extracted by water extraction method from dark tea.Develop handmade soap and facialcleanser with dark tea functional ingredients.The amount of total soap,total fat and dry sodium soap content of hand soap is positively correlated with the amount of dark tea added.The in vitro moisturizing properties of facial cleanser are positively correlated with the amount of dark tea added.When lower temperature and greater humidity,moisturizing will be better.The removal rate of DPPH free and hydroxyl radicals improve significantly with functional ingredients of dark tea in facial cleanser.And the removal rate of DPPH free is more significantly.
