Cordyceps,including Cordyceps kyushuensis and Cordyceps militaris,has good medicinal value.The cordycepin,polysaccharide and other substances produced by these have function of antitumor,antioxidant,hypoglycemic and so on.Radix Astragalus,Ganoderma lucidum,Scutellaria barbata and Pan ax ginseng C.A.Mey are common traditional Chinese medicines,whose active ingredients are complex and diverse,and they also have anti-tumor and anti-oxidation effects.By solid-state fermentation of traditional Chinese medicine and Cordyceps,the medicinal efficacy can be improved.However,what changes in Cordyceps metabolism after adding traditional Chinese medicine into the culture medium remains to be explored.In this study,Cordyceps kyushuensis and Cordyceps militaris were selected for solid-state fermentation with Astragalus,Ganoderma lucidum,Scutellaria barbata and Panax ginseng C.A.Mey.Firstly,Cordyceps kyushuensis and Cordyceps militaris were inoculated on four kinds of traditional Chinese medicine-rice medium respectively for solid fermentation,and the content of cordycepin and other active substances were detected.It was found that the content of cordycepin could be increased by adding traditional Chinese medicine.The fermentation effect of Cordyceps kyushuensis was better than that of Cordyceps militaris,and the content of cordycepin in Radix Astragalus medium was the highest,followed by Ganoderma lucidum medium.Then,adjusted the proportion of Radix Astragalus and Ganoderma lucidum in the medium(10%,20%,30%,40%and 50%),inoculated with Cordyceps kyushuensis,detected and compared the content of cordycepin,pentostatin and polysaccharide.The results show that the content of active substances in Radix Astragalus group is higher than that in Ganoderma lucidum group,while the dosage of traditional Chinese medicine is more than 20%,the content of the above-mentioned substances was higher in the fermentation.In addition,the toxicity of the fermented products of 20%traditional Chinese medicine on zebrafish embryo development was evaluated.It was not found that the Radix Astragalus group had an effect on zebrafish embryo development,while the Ganoderma lucidum group had a certain toxic effectAfter that,20%Radix Astragalus was added into rice culture medium to culture Cordyceps kyushuensis.The fermentation were collected in the mycelium stage(CKK1),coloring stage(CKK2),stromata-forming initial stage(CKK3)and fruiting body stage(CKK4)of Cordyceps kyushuensis and the dynamic changes of flavonoids,saponins,cordycepin,adenosine and other active substances content were detected.The results showed that the content of adenosine and cordycepin increased first and then decreased with the increase of fermentation time,and the content was highest at the 30th day of fermentation.The flavonoid content gradually decreased.The content of saponins increased first and then decreased.On the other hand,the transcriptome of four different growth stages of Cordyceps kyushuensis was sequenced,Transcriptome sequencing analysis found that in CKK1,the expression of genes regulating hydrophobin synthesis was higher than the other three stages,which was conducive to mycelial growth.In CKK2,the expression of genes related to purine metabolism was higher than the other three periods,so more adenosine and cordycepin were synthesized.In CKK3,a large number of substances such as lipids and aromatic amino acids were synthesized,and at the same time,intracellular information transmission was increased.In CKK4,nucleic acid metabolism,ribosome formation,and carbon metabolism accelerated,and cells proliferated to form fruiting bodies and spores.
