A variety of methods could be used to detect carbohydrates in food because of different types and content.Detection of carbohydrates for aquatic products should be established,due to the special nature of raw materials and processing methods.As a major category of food,detection method of carbohydrates in aquatic products is the important basis of determining its quality.The establishment of carbohydrates determination method is the prerequisite for the quality control of aquatic products.In this paper,the method of detecting the total sugar content of aquatic products was optimized,and the method of analyzing the content of monosaccharide in sea cucumber was established.It analyzed the composition and content of monosaccharide in sea cucumber and its products,which provided the theory of sugar-related standards in aquatic products.It analyzed the influence of processing methods on the nutrient composition of sea cucumber.Research contents are as follows:In this paper,the method of detecting total sugar content of aquatic products was established content of total sugar in aquatic products was detected by phenol-sulfuric acid with visible spectrophotometer.The effect of hydrolysis conditions on the total sugar content was analyzed by single factor experiment and the optimal hydrolysis conditions of total sugar were determined by L9(34)orthogonal test.Hydrolysis conditions were optimized for example,hydrochloric acid concentration,hydrolysis time and hydrolysis temperature,and were verified by measuring the total sugar content of the sample.The results of single factor experiments show that the hydrolysis conditions affect the total sugar content of aquatic products.The results of orthogonal test show that temperature is the most important factor affecting the hydrolysis conditions.The appropriate hydrolysis conditions of total sugar in aquatic products were as follows: hydrochloric acid concentration of 20%,hydrolysis time of half an hour,and hydrolysis temperature of 70 ?.The linear range of this method is between 0 and 100 ?g/mL,and the relative standard deviation is 0.8% with good reproducibility.The average recoveries of shredded squid,dried abalone and dried scallop adductor were between 92.3% and 117%.Phenol-sulfuric acid method has high accuracy and precision,which is suitable for the detection of total sugar content in aquatic products.In this paper,a high performance anion exchange-pulse amperometric assay(HPAEC-PAD)was used to analyze the composition and content of polysaccharides in sea cucumber polysaccharides.A method has been developed for determination of monosaccharide composition and content in sea cucumber polysaccharide by HPAEC-PAD.Method of purifying sea cucumber polysaccharide was optimized by comparing three kinds of deproteinization methods.Optimum chromatographic conditions for the separation of monosaccharide were optimized by means of comparing the effects of eluent concentration on the separation of monosaccharide.The results indicated that reagent of sevag could remove micro-molecule polypeptide more effectively than others from sea cucumber polysaccharides.The concentration of eluent had significant influence on separating monosaccharide.Eight kinds of monosaccharide,like Fuc?Gal?Ara?GalN?GlcN?Man?Glc?Lac were separated under the condition of using 20 m M Na OH as eluent.GlcUA and Gal UA were separated by using 160 mM NaOH and 200 m M NaAC as eluent.But the speed of mobile phase had minute affect on the separation of monosaccharide.The linear coefficients of monosaccharide standard were above 0.998.The detection limit of each monosaccharide was between 2.5 and 50 ?g/L.The recoveries ranged from 83.6 to 113.1% and the relative standard deviation was between 0.06% and 3.81%.The method could determine neutral sugar,amino sugar and uronic acid in sea cucumber polysaccharides,which could be used as analysis and quantitative method of monosaccharide in sea cucumber and its pr oducts.The composition and content of monosaccharide were significant indexes for determining the biological activity of sea cucumber.The establishment of method could provide basic monosaccharide information and promote the polysaccharides research progress in sea cucumber.It could provide references for the establishment of sea cucumber standards,and expand the application of active substances in medicine and other fields.The composition and content of monosaccharide was analyzed in seven kinds of sea cucumbers by HPAEC-PAD,like Apostichopus japonicas? Cucumaria japonica? Stichopus variegatus? Holothuria nobilis?Parastichopus californicus?Isostichopus fuscus and Isostichopus badionotus.The composition of monosaccharide was different in different types and places of origin in sea cucumber.It was analyzed of the composition of monosaccharide in sea cucumber dried powder by HPAEC-PAD.The results showed that the content of monosaccharide was rich in sea cucumber internal organs.The composition analysis of sea cucumber monosaccharide provided guidance for the research of biological activity and nutritional value of sea cucumber.The determination of sea cucumber monosaccharide by HPAEC-PAD provides technical support for the analysis of the nutritional value and quality of sea cucumber,and further improves the quality standard system of sea cucumber.Effects of processing methods on nutrient composition of sea cucumber were analyzed by detecting chemical indicators,like protein,polysaccharide and monosaccharide.The traditional processing method had different degrees of loss.Boiling would reduce the polysaccharide content of sea cucumber instead of protein.Salting and desalting repeatedly could reduce the content of polysaccharide and protein significantly.It could provide reference for the research and development of new products of sea cucumber,by means of analyzing the effect of processing methods on the nutrient composition of sea cucumber.
