The Study Of Zhiyangxiaoyan Solution For Mosquito Bites

Insect bite dermatitis is acute inflammatory skin disease,caused by insect bites or pseudoparasitism and poison seta stimulation.It is a common disease in department of dermatology in summer,and the main clinical manifestations are local skin rash and pruritus.With high incidence and skin itching,insect bite dermatitis affect daily life and the rest of the people.The conventional treatment methods of insect bite dermatitis are as follows: topical glucocorticoid preparation and topical antipruritic agent such as calamine lotion for mild symptoms,oral antihistamines for extensive lesions,oral corticosteroids in short term for severe allergic reaction and antibacterial drugs for secondary infection.The corticosteroid cream is more common than other topical preparations for insect bite dermatitis in the current market.Its characteristic is the anti-inflammatory effect is obvious,while the antipruritic effect is poor.Therefore,this study intends to develop a kind of topical liquid preparation with anti-inflammatory,antipruritic and anti-allergic effect,which can be used to cure insect bite dermatitis to relieve itching symptoms and eliminate rash.The preparation take the antihistamine diphenhydramine hydrochloride,the glucocorticoid dexamethasone acetate and the anti-inflammatory drug enoxolone as primary medicines,combined with camphor and menthol.The dexamethasone acetate,diphenhydramine hydrochloride and enoxolone in zhiyangxiaoyan solution were analyzed by high performance liquid chromatograph(HPLC),with XDB-CN column,the mobile phase consisted of methanol-1%triethylamine(the pH was adjusted to 6.5 with glacial acetic acid)(49.5:50.5).This analysis method is specific,sensitive and simple.It can meet the demands of content determination with zhiyangxiaoyan solution.With the stability and percutaneous permeability as the evaluation index,the prescription and preparation process of mosquito bites zhiyangxiaoyan solution were optimized and determined by the single factor exploration.The final prescription was:0.025% of dexamethasone acetate,2.0% of diphenhydramine hydrochloride,0.2% ofenoxolone,1.0% of camphor,3.5% of menthol,0.1% of 3-Methyl-4-isopropylphenol,0.02% of butylated hydroxytoluene,0.5% of citrate,0.8% of sodium citrate,5.0% of purified water,10.0% of 1,2-propylene glycol,and 76.855% of 70% ethanol.The mosquito bites zhiyangxiaoyan solution is colorless transparent liquid,with the scent of camphor and menthol,pH 4.5-5.5.The cumulative permeation quantity in 24 h of dexamethasone acetate,diphenhydramine hydrochloride and enoxolone in zhiyangxiaoyan solution was 30.91 ?g·cm-2,3368.86 ?g·cm-2 and 131.55 ?g·cm-2.The steady penetration rate of dexamethasone acetate,diphenhydramine hydrochloride and enoxolone in zhiyangxiaoyan solution was 1.81 ?g·cm-2·h-1,194.05 ?g·cm-2·h-1 and 7.75?g·cm-2·h-1.The efficacy of the mosquito bites zhiyangxiaoyan solution was investigated by the histamine phosphate induced guinea pig itching test,the xylene induced mouse ear swelling test and mouse hot plate nociceptive test.In the histamine phosphate induced guinea pig itching test,zhiyangxiaoyan solution can significantly inhibit itching duration,with the inhibition rate of 99.78%(P<0.01).The antipruritic effect of zhiyangxiaoyan solution was comparable with mopidick lotion and compound camphor cream,and was superior to compound dexamethasone acetate cream(P< 0.01).In the xylene induced mouse ear swelling test,zhiyangxiaoyan solution can significantly inhibit the ear swelling degree,with the inhibition rate of 62.28%(P<0.01).The anti-inflammatory effect of zhiyangxiaoyan solution was comparable with mopidick lotion,and was better than compound camphor cream(P<0.01)and compound dexamethasone acetate cream(P<0.05).In mouse hot plate nociceptive test,zhiyangxiaoyan solution can increase the pain threshold of mice to a certain extent,with the increase rate of 22.00%(P<0.01).Pharmacodynamics study showed that self-made mosquito bites zhiyangxiaoyan solution combined with dexamethasone acetate,diphenhydramine hydrochloride and enoxolone,has a good antipruritic,anti-inflammatory and anti-allergic effects.Impact factor test,accelerated test and long-term test were applied to observe the stability of mosquito bites zhiyangxiaoyan solution.The result of impact factor tests showed that self-made mosquito bites zhiyangxiaoyan solution is sensitive to hightemperature and strong light,should be protected from light and shade storage.In the 6months accelerated test and long-term test,the self-made zhiyangxiaoyan solution had no significant changes in the indexes,and its quality is stable.Currently,the long-term test is still in progress.To establish the quality control method of mosquito bites zhiyangxiaoyan solution,provides good quality control and evaluation method to ensure the quality of the preparation.The prescription and preparation process of mosquito bites zhiyangxiaoyan solution is reasonable and feasible,and its quality is stable with good curative effect.
