Ion chromatography(IC)has the advantage of rapid analysis,high efficiency,high sensitivity and simple operation.It has wide application in the modern analysis field,such as food sanitation,environmental safety,chemistry engineering,biology engineering and so on.Ion chromatography is a liquid chromatography that based on the object can be dissociable and the stationary phase surface has the charged functional groups interact,so that the object remains on the stationary phase.Due to the different of acting force between the stationary phase and different ion,the separation of object is achieved when the object flows successively the column.These years,the pharmacologic action of carbohydrate compounds were came to light and some carbohydrate compounds can be regard as the significant gist to measure the quantity of products.Therefore,analysis of carbohydrates which is the plenty of researchers chase after.IC to analysis of carbohydrates has been identified as an erective method.It plays an important role in the food industry.The paper mainly includes two parts: review and research report.In the first part,a review includes the development and concept of ion chromatography,the classification and principle of IC and the detection technology at first.Secondly,the application of IC in food analysis was expound.At last,it briefly introduced the carbohydrate compounds which include the concept and classification,the extraction and the analytical methods.In the research report part,the extraction and separation method of carbohydrate compounds in tropical fruits,flowers,Chinese dates and ready-to-drink green tea was established.1.Determination of glucose,sucrose and fructose in 10 kinds of fruits by ionchromatographyA high performance anion exchange-pulsed amperometric detection(HPAE-PAD)chromatographic method was applied to determine glucose,sucrose and fructose in pitaya,wax apple,avocado and other 7 kinds of tropical fruits.After ultrasonic extraction,the polysaccharide in 10 kinds of fruits was separated by METROSEP CARB 1(150 mm×4.0 mm)anion exchange column and detected by pulsed amperometric detector.30.0 mmol/L NaOH was used for isocratic elution.Analysis of the samples could be finished within 18 min.Detection limit of glucose and sucrose were 0.1209 mg/L and 0.2827 mg/L respectively.The linearity ranged from 1.0 mg/L to 70.0 mg/L.Detection limit of the fructose was 0.5501 mg/L.The linearity range was 5.0~80.0 mg/L.The RSDs of 3 polysaccharide contents for 5 consecutive determinations were between 0.67% and 7.04%,and the spike recoveries were between 78.70% and 117.75%.The method has the characteristics of simple pretreatment,good selectivity and high sensitivity,and it can be applied in the determination of soluble sugars in fruits.2.Ultrasonic assisted extraction of free sugar in 5 kinds of flowers by ion chromatographyThe IC method based on ultrasonic assisted extraction was developed for separation and determination of free sugars(galactose,glucose,mannose and fructose)in lily,sweet osmanthus,chrysanthemum,honeysuckle and rose.After ultrasonic extraction at room temperature,the separation of galactose,glucose,mannose and fructose was achieved on METROSEP CARB 1(150 mm×4.0 mm)anion exchange column and detected by pulsed amperometric detector.8.0 mmol/L NaOH was used for isocratic elution.Detection limit of galactose,glucose and mannose were 0.2275 mg/L,0.3452 mg/L and 0.4798 mg/L respectively.The linearity ranged from 1.0 mg/L to 60.0 mg/L.Detection limit of the fructose was 1.9103 mg/L.The linearity range was 2.0-70.0 mg/L.The RSDs of peak height for 6 consecutive determinations of 10.0 mg/L galactose,10.0 mg/L glucose,10.0 mg/L mannose and 20.0 mg/L fructose were 2.66%,2.81%,2.12% and 4.80%,respectively.The RSDs of peak area were 3.23%,5.41%,4.19% and 7.03%,respectively.The RSDs of retention time were 1.92%,2.64%,3.01% and 4.62%,respectively.The spike recoveries were between 72.31% and 112.0%.The method has the characteristics of simple pretreatment,good selectivity and high sensitivity,and it provides experimental proof for separation and assaying of free sugars in flowers.3.Alkaline liquor extraction of three kinds of sugar in Chinese Dates by Ion ChromatographyAlkaline liquor was used to extraction solvent,the HPAE-OAD chromatographic method for assaying of three kinds of sugar(glucose,fructose and sucrose)in Chinese dates.The separation was obtained on METROSEP CARB 1(150 mm×4.0 mm)anion exchange column and detected by pulsed amperometric detector.180 mmol/L NaOH was used for isocratic elution.Analysis of the samples can be finished within 8 min.The extraction conditions was alkaline liquor extracted by 1:20(g: mL)liquor ratio of 0.5 mol/L Na2CO3 for 20 min at 90 ?.Detection limit of glucose,fructose and sucrose were 0.0682 mg/L,0.1775 mg/L and 0.1910 mg/L separately.The linearity range of are 1~60 mg/L.The relative standard deviation was between 1.22% and 5.35%(n=5).The recoveries are between 82.15% and 120.65%.The method has good selectivity,high level of sensitivity and was suitable for determination of glucose,fructose and sucrose in Chinese dates.4.Determination of monosaccharide and oligosaccharide in 5 kinds of ready-to-drink green tea by anion exchange chromatography with pulsed amperometric detectorThe HPAE-PAD chromatographic method was developed for separation and determination of monosaccharide and oligosaccharide(sucrose,glucose,mannose and fructose)in 5 kinds of ready-to-drink green tea.The separation of sucrose,glucose,mannose and fructose was achieved on METROSEP CARB 1(150 mm×4.0 mm)anion exchange column with isocratic elution.Mobile phase was 7.0 mmol/L NaOH,and flow rate was set at 1.0 mL/min.The simultaneous separation of four kinds of sugar was obtained within 30 min,and satisfactory linear equations(r>0.998 0,n=8)were established.Detection limit of sucrose,glucose,mannose and fructose were 0.2995 mg/L,0.1144 mg/L,0.1842 mg/L and 1.1642 mg/L separately.The linearity range of sucrose,glucose and mannose are 1.0~60.0 mg/L.The linearity range of fructose are 5.0~80.0 mg/L.The average recoveries of real sample were between 87.00% and 107.60%.The intraday precision of sucrose,glucose,mannose and fructose were 1..25%,0.68%,2.57% and 3.74% separately,and the daytime precision were 3.56%,2.09%,4.63% and 5.87% respectively.The method was environment-friendly,accuracy and thus can provide a reference for quality control of ready-to-drink green tea.
