Undaria pinnatifida can be used as food and medicine,which is one of the main economic marine algae.Undaria Pinnatifida also has high nutritional value.Undaria pinnatifida sporophyll has hard texture,it is often discarded in the processing.The pigment of lutein is a kind of lutein,which is a kind of pigment in lutein,which has a variety of physiological functions,it is a very important raw material in the Undaria pinnitafida.In order to solve the waste of the undaria pinnatifida sporophyll,to improve the utilization rate of it,and use fucoxanthin for many applications,we choose the undaria pinnatifida sporophyll as the research object,and research on two aspects.On the one hand,the method of ultrasonic extraction,enzymatic extraction and aqueous two-phase methods were used to prepare fucoxanthin.In order to determine the optimal extraction process,the extraction solvent,material liquid ratio,ultrasonic time,enzymolysis time,enzymolysis temperature and the ratio of salt and alcohol are screened.On the other hand,we studied fucoxanthin physiological activity.The free radical scavenging ability of DPPH and OH in vitro and the effect on the activity of lipase and amylase are investigated.In addition,the antioxidant ability of fish oil and pork was studied.At the same time by using electron spin resonance(ESR)spectroscopy to investigate the effect of fucoxanthin on inhibiting free radical during food lipid peroxidation models,which include fish oil,corn oil,linoleic acid and linoleic acid salt 4 food lipid peroxidation models.To study the protective effect of fucoxanthi on RAW 264.7 cells induced by H2O2.When complex processes including ultrasonic method and enzymatic hydrolysis was used to extract fucoxanthin.The results indicated that the best extraction amount(0.75±0.03 mg/g)could be reached under the following conditions:ratio of ethanol and acetone was 3:1,ratio of solid and liquid was 1:40(w:v),ultrasonic time was 30 min,enzyme hydrolysis time was 35 min,temperature was 50 ?.When ethanol-phosphate aqueous two-phase system was used to extract fucoxanthin.The results indicated that the best extraction amount(2.65±0.21 mg/g)could be reached under the following conditions:ratio of solid and liquid was was 1:20(w/v),phosphate(w/w),20%ethanol 25%(w/w),extraction temperature was 40 ?,extraction time was 2h.The result indicated that the DPPH radical and hydroxyl radical scavenging ability enhanced with the increasing concentration of fucoxanthin,and IC50 of DPPH radical scavenging was 3.9 p.g/mL,IC50 of hydroxyl radical scavenging was 34.9 ?g/mL.The inhibition rates of lipase and alpha amylase were 54.33±0.42%and 29.52±4.38%,respectively.The results showed that fucoxanthin could effectively inhibit the production of MDA in fish oil and pork model.And the highest inhibition rate was 78.1%and 80.9%.Electron spin resonance spectroscopy(ESR)was used to detect the formation of lipid free radicals in 4 food models,when fucoxanthin concentration reached 100 ng/mL,accelerated oxidation for 3 h,inhibition of lipid free radical rate were 64.46±6.46%and 57.4±5.15%in fish oil and corn oil model.When fucoxanthin concentrations reached 300 ng/mL,accelerated oxidation for 10 min,inhibition of lipid free radical rate were 75.24%and 66.9%in linoleic acid and linoleic acid salt model.Studies have shown that the fucoxanthin can effectively prevent the oxidation of RAW 264.7.The content of ROS decreased 27.5%,the content of MDA decreased 39.1%,the activity of SOD increase 123.2%,the activity of CAT increase 43.3%,and the activity of GSH-Px increase about 152.5%To solve the problem of low utilization of by-product through study on the process parameters that extraction of fucoxanthin from undaria pinnatifida sporophyll.And provide a new way to obtain fucoxanthine.At the same time,it provides a theoretical basis for its application in the field of health food and food antioxidants.
