Uncontrolled hemorrhage is a leading cause of prehospital death after military and civilian trauma,so effective control of blood loss could be the primary action to reduce the mortality.Rapid and effective hemostasis is the necessary guarantee for the safety of patients after traumatic surgery.To investigate the application of silk fibroin to hemostatic material,the fabrication and properties were researched on the composite among silk fibroin,gelatin and calcium alginate,and the water-absorbing quality,compressibility,clotting hemorrhagic and cytocompatibility of composite were conducted.Firstly,the properties of silk fibroin/gelatin composite porous materials were studied.The results show that the water absorption of the composites gradually increased and the softness became worse with the increasing of the proportion of gelatin.When the ratio of silk fibroin/gelatin was 75:25,the water absorption of the material reached the highest,the rate of instantaneous water absorption and the ultimate water absorption reached the maximum?about 1021%and 1118%?,increased by 15%and 20%,respectively.SEM results indicated that the porosity of the composites became bigger with the increasing of gelatin ratio.The APTT,PT and TT of normal blood was 35.62s,20.25s and 26.23s,respectively,and those of 50:50 group was 26.73s,16.62s and 19.13s,reduced by 25%,18%and 25%,respectively.The50:50 group in the coagulation effect was significantly improved and the results were similar to those of the whole blood.The results of fibrinogen showed that the content of fibrin on the material increased with the increasing of gelatin ratio in the composites,when the ratio reached 50:50,the fibrin content of the material increased significantly.The results of cytocompatibility suggested that the cells grew well in the composites compared with the control?TCP plate?after 5 days,and the number and morphology of the cells had no significant difference,which indicated that the composites had a good cytocompatibility.Secondly,the properties of silk fibroin/calcium alginate composite porous materials were studied.The results showed that the water absorption of the composites increased at the beginning and then decreased with the increasing of the proportion of calcium alginate.When the ratio of silk fibroin/calcium alginate was 75:25,the water absorption of the material reached the highest,the instantaneous water absorption and the ultimate water absorption were 2033%and 2267%,respectively.Compressibility experiments showed that the softness of the material decreased with the increasing of the proportion of calcium alginate in the material.The SEM results showed that the porosity of the materials had no significant difference with the changing of the proportion of calcium alginate.The APTT,PT and TT of normal blood was 32.83s,19.26s and 24.51s,respectively.The 50:50 group was 28.13s,14.49s and 14.26s,reduced by 25%,18%and 25%,respectively.The clotting time of the material was significantly reduced.The whole blood testing results showed that the coagulation effect of 50:50 group was better than that of other groups.The fibrinogen results showed that the content of fibrin on the material increased with the increasing of the proportion of calcium alginate in the composites.The results of cytocompatibility suggested that the cells grew well in the composites compared with the control?TCP plate?after 5 days,and the number and morphology of the cells had no significant difference,which indicated that the composites had a good cytocompatibility.Finally,the properties of silk fibroin/gelatin/calcium alginate composite porous materials with a mass fraction of 3%were studied.The results of water absorption showed that the instantaneous water absorption and ultimate water absorption of the mass ratio of1:1:1 are 2.3 times and 1.6 times than the mass ratio of 1:1:3,respectively.Compressibilit experiments showed that the mass ratio of 1:1:3 is about 1.8 times than the 1:1:1 material,the mass ratio of 1:1:1 is better in flexibility;The whole blood testing,APTT,PT,TT results showed that there were no significant differences in the clotting time between those two groups;The results of platelet adhesion test showed that 5min and 10min,the number of 1:1:1 at OD340 is about 2 times than that of 1:1:3.The mass ratio of 1:1:1 was batter in platelet adhesion.The fibrinogen results showed that the fibrin content of 1:1:1 is 3 times than the material of 1:1:3;The results of cytocompatibility suggested that the cells grew well in the composites compared with the control?TCP plate?after 5 days,and the number and morphology of the cells had no significant difference,which indicated that those two composites had a good cytocompatibility.
