Southwestern region of China has a vast karst landform feature.Karst water is the main source of water for industrial and residential life in the area.However,the phenomenon of the garbage accumulation and the sewage disorderly rowing is prominent,which seriously pollutes the groundwater system in the area.Due to the complex terrain conditions,the key to protect the environment and control the pollution is how to quickly and efficiently detect the groundwater pollution in the area and explore its source of pollution.This article will explore the practicality of the microbial source tracer technology through on-site investigations,sampling and detection,combined with high-throughput sequencing,microbial diversity,and primer-specific analysis methods,so as to lay a foundation for follow-up the use of contaminated waters and microbial source tracking methods for pollution quickly tracking.The main research contents and results of this paper are as follows:(1)There was significant antibiotic contamination in the study area and the abundance of antibiotic resistance genes(ARGs)was high,and there was a positive correlation with antibiotic concentrations.(2)In the karst water system,the distribution of microbial communities is closely related to the environmental factors,among them,Proteobacteria and Bacteroidetes are the dominant bacteria,which can be used to screen the main indicator bacteria traced by microbial sources,so as to design more efficient and sensitive primers for pollution sources tracking.(3)Through PCR sensitivity and specificity experiments,we choose SulII,ermF,BlaOXA-1,and rpoC genes can effectively utilize for microbial source tracking studies,through karst groundwater river flow,environmental factors,microbial community analysis,and microbiology source tracking were used to judge that the source of pollution in the study area(mainly antibiotic contamination)comes from the discharge of WWTP.
