ObjectiveTo set up the antibiotic resistance analysis database of Nanjing Zhongshan River, track the sources of the pollution and evaluate the result.MethodsFecal samples, industrial and domestic waste water samples were collected around Nanjing Zhongshan River. Indicator bacteria isolates were isolated from these samples. Tryticase soya agar(TSA) plates containing antibiotics with different concentrations were prepared. Isolates were transfered onto the TSA plates and scored by the growth. Microbial source tracking database were bulit by ARA experiment and were evaluated by the rate of correct classification. Water samples were taken from Zhongshang River according to the plan. Indicator bacteria isolates were isolated by membrane filtration methord. Then, ARA experiments were done. At last, pollution sources were tracked and evaluated by discriminant analysis using JMP7.0 software.Results(1) The known-source ARA library contained 2792 known source E. coli: 865 isolates from human, 315 isolates from pig, 237 isolates from cattle, 117 isolates from sheep, 331 isolates from chicken, 305 isolates from duck, 48 isolates from goose, 360 isolates from dog, 214 isolates from sewage samples.(2) The known source library was divided into four parts (spring, summer, autumn, winter part) to analysis. In spring part, the ARCC is 70.61% when the library is classified into three categories(human, animal, sewage), 60.27% when classified into five categories(human, poultry, livestock, dog, sewage); in summer part, the ARCC is 68.75% when the library is classified into three categories, 52.47% when classified into five categories; in autumn part, the ARCC is 62.52% when the library is classified into three categories, 48.21% when classified into five categories; in winter part, the ARCC is 69.57% when the library is classified into three categories, 62.77% when classified into five categories.(3) In spring, the major pollution source of Zhongshan river is sewage. In summer, the major pollution source of Zhongshan river is feces of dog. In autumn, the major pollution sources of Zhongshan river are sewage and feces of human. In winter, the major pollution sources of Zhongshan river are feces of human and dog.(4) Compare with the result from the ARA study in Guiwu reservoir of the Huai River, the ARCCs from this study are lower.ConclusionThe antibiotic resistance analysis as a microbial source tracking method is available for both ecosystem water body and dual polluted water body.
