The instrumentation of analytical tools and complication of analytical objects provide the new opportunities for the researchers of analysis, and also bring out a huge challenge, that how to extract more useful chemical information from a large number of raw measured data.Chemometrics is a powerful tool for handling the complex multiway data, and has been successfully applied in analytical chemistry and other related fields.Second-order calibration method is the one of the hotspots of chemometrics in recent years, based on the existence of the "second-order advantage",it can achieve simultaneous, accurate quantitative analysis of multiple analytes of target in complex systems even in the presence of unknown interferences. From reality, combining the second-order calibration method with different instruments, the author carries out the related research work about the cosmetic and food analysis. The main contents are as follows:In Chapter 2,Arbutin and hydroquinone are two tyrosinase inhibitors and often used in skin care products as whitening agents. In this work,a practical analytical method that combines the excitation-emission matrix fluorescence with second-order calibration method based on alternating trilinear decomposition(ATLD)algorithm was proposed for simultaneous determination of arbutin and hydroquinone in cosmetics. This method successfully achieved quantitative analysis of arbutin and hydroquinone in three different complex systems simultaneously, even in the presence of unknown, uncalibrated interferences.Besides, the method was also validated by high performance liquid chromatography(HPLC),and the results obtained by the two methods were compared by t-test and F-test and were found to be in good agreement with each other, but this method was environmentally friendly and easy.So the developed method can be an alternative method for the determination of arbutin and hydroquinone simultaneously and can be used to quality monitoring of whitening cosmetics. In Chapter 3,An attractive strategy that combines the second-order calibration method based on alternating trilinear decomposition algorithm(ATLD)with high performance liquid chromatography with diode array detector(HPLC-DAD)was developed to mathematically separate the overlapped peaks and to rapidly quantify four furocoumarins in cosmetic products. This new strategy proved to be a useful tool to handle the co-eluted peaks and uncalibrated interferences existing in the process of chromatographic separation, which benefited from the "second-order advantage",making the determination of furocoumarins within 4.0 min under a simple mobile phase condition possible. This method shows good linearity(R2>0.99),rapidity and accuracy, which may be extremely promising as an excellent routine strategy for identification and quantification of furocoumarins in the complex matrices. In Chapter 4,Both sesamin and sesamolin are characteristic lignans of sesame oil, their presence greatly improves the stability of sesame oil,as well as the material basis for the sesame oil has many physiological health function.Considering that sesamin and sesamolin are natural fluorophores,a sensitive three-dimensional fluorescence method was proposed for simultaneous determination of sesamin and sesamolin in sesame oil with the aid of the second-order calibration method based on alternating trilinear decomposition algorithm(ATLD).Even the fluorescence spectra of analytes of target have the serious high collinearity,by the "mathematical separation" of second order calibration method to replace or enhance conventional"physical or chemical separation",making the simultaneous and direct quantitative analysis of analytes of target possible. The developed method aviods laborious sample pretreatment processes, and would be of great help to sesame oil industries in monitoring their quality. In Chapter 5,The abuse of oral antidiabetic agents brings out many side effects,so the quantitative analysis of these kinds of drugs in human plasma and"antidiabetic" health products has very important practical significance.A novel chemometrics-enhanced high performance liquid chromatography with diode array detector(HPLC-DAD)method was developed to simultaneous and fast determination of eight oral antidiabetic agents in human plasma and "antidiabetic" health products.A few co-eluted peaks allow one to shorten the time needed to accomplish the chromatographic analysis in less than 9.5 min.Compared with the traditional method, the analysis efficiency was improved, the use of organic solvents was reduced, satisfactory quantitative results were obtained even in the presence of co-eluted peaks and unknown interferences.
