Phellinus linteus belongs to one of perennial large medicinal fungi.Polysaccharide is one of the main bioactive components of Phellinus linteus.It has antitumor,bacteriostatic,enhancing organism immunity,reducing blood sugar and other biological activities.At present,there are many reports about the fungal polysaccharide,but less research on the polysaccharide of Phellinus linteus.This paper aims to discuss the extraction methods of Phellinus linteus polysaccharide and its antibacterial activity to provide basis for its future in depth development and utilization.The extraction of Phellinus linteus polysaccharides and purification technology are the foundations of the further use of Phellinus linteus polysaccharides.In this paper,Phellinus linteus mycelium of Taishan was used as raw material,the modern food separation technology,food processing technology and modern instrumental analysis techniques were used in Taishan Phellinus linteus mycelium polysaccharide extraction Research.The polysaccharides of Phellinus linteus mycelium were separated and purified by DEAE-52 Cellulose column chromatography and Sephadex G-100 column chromatography,and the antibacterial activities of the three polysaccharides were preliminarily discussed.The main findings are as follows:(1)The optimum extraction technology of hot water extraction was determined by orthogonal experiment as follows: the particle size of 60 mesh,the extraction temperature was 90?,the extraction time was 2.5 h,the ratio of extraction liquid to solid was 30:1 g/g,two times.Under these conditions,polysaccharide yield was 2.88%.(2)On the basis of the optimum technology of hot water extraction,the optimum extraction process of microwave pretreatment-hot water extraction was determined by orthogonal experiment as follows: microwave pretreatment time was 5 min,power was 500 W,liquid-solid ratio was 20:1 g/g,the particle size of hot water extraction was 60 mesh,the extraction temperature was 90?,the extraction time was 2.5 h,the ratio of extraction liquid to solid was 30:1 g/g,two times.Under these conditions,polysaccharide yield was 5.15%.(3)Under the optimum condition of microwave pretreatment,the optimum extraction process of microwave pretreatment-ultrasonic-assisted enzyme extraction was obtained by the design of Plackett-Burman test and response surface test as follows: microwave pretreatment time was 5 min,power was 500 W,liquid-solid ratio was 20:1 g/g,the liquidsolid ratio of ultrasonic assisted composite enzyme was 20:1 g/g,extraction time was 22 min,extraction temperature was 49?,extraction power was 420 W,6.4 pH,cellulase dosage added was 0.7%,protease dosage added was 0.9%,two times.Under the conditions of this process,the polysaccharide yield of Phellinus linteus was 6.84%,the extraction rate of polysaccharides was 91.57%.(4)The components of 3 polysaccharides were extracted by column chromatography,they were Php1-1,Php1-2,Php2-1,respectively.The three polysaccharides had different antimicrobial activities,and the bacteriostatic effect of Php1-2 was better than that of Php1-1 and Php2-1.The three polysaccharides had no inhibitory effect on Candida albicans and Aspergillus Niger in the concentration range of 0 ~ 80 mg/m L.The experimental results show that the polysaccharide of Taishan Phellinus linteus has good inhibitory effect on bacteria.
