The optimum conditions for isolation,purification of polysaccharides from mycelium of Gloeostereum incarnatum were studied in this paper.And then got the GⅠ-Ⅱc which was purified from polysaccharides.The physicochemical properties and component of the polysaccharides from mycelium of Gloeostereum incarnatum have been identified.The result of research shows that it has distinct antioxidant activity.Use ethanol grade precipitation to extract the polysaccharides.Choose the fractions were precipitates with 70%alcohol named GⅠ-Ⅱas subject of study which yield was 2.61%.It has found that the optimum deproteinzation condition by orthogonal tests used the method of Sevag was as follows:Vsample:Vchloroform+n-butanol=3:1,Vchloroform:Vn-butanol = 5:1,shaking time was 15 minutes and 2 times.Decolorizing experiments used AB-8 resin.The best conditions for de-colorization is that AB-8 resin 0.2g/mL,pH 4.0,T=60℃.GⅠ-Ⅱ were further graded and purified by DEAE-cellulose 52 and get three fractions named GⅠ-Ⅱa,GⅠ-Ⅱb,GⅠ-Ⅱc.GⅠ-Ⅱc was a pure polysaccharide.The purity was 95.2%.The study of physicochemical properties was showed that all of them were non-starch polysaccharide without polyphenol,protein and nucieic acid.The analysis of IR spectrum shows GⅠ-Ⅱa,GⅠ-Ⅱb and GⅠ-Ⅱc possessed the characteristics of carbohydrate absorption peak which in the form of pyranose.GⅠ-Ⅱc contained the composition of amide.The analysis of TLC indicated both GⅠ-Ⅱ a and GⅠ-Ⅱb were made up of Glu,Gal,Xyl,Man.But GⅠ-Ⅱc maybe was only made up of Glu.The result of antioxidant activities in vitro of polysaccharides from Gloeostereum incarnatum shows:GⅠ-Ⅱ has distinct antioxidant activity especially in ability of reducing capacity and hydroxyl and DPPH radicals scavenging activities.0.1mg/mL GⅠ-Ⅱ’s reducing capacity was as long as Vc’s 75%.The effect of scavenging hydroxyl radicals of GⅠ-Ⅱ was much better than Vc,IC50=0.124mg/mL.It can also scavenging DPPH radicals which IC50=58.82 μg/mL.But the superoxide radicals scavenging activities of the polysaccharides were unideal.In the three fractions of polysaccharides,GⅠ-Ⅱc was the component which has the best antioxidant activities.Through anti-biotic experiment,it showed that the.large molecular polysaccharides don’t have anti-biotic capacity.
