This paper takes the benzopyrene,a common PAH contaminant in the ocean as the target contaminant,and the Sebasticus marmoratus,a common small-scale benthic burrowing fish in Chinese ocean as the experimental subject.Injecting toxicity experiments were conducted to study the toxic effects of different concentrations and different times on the liver of S.marmoratus to test the concentration of benzo[a]pyrene that makes the CYP1A1 m RNA gene expression of S.marmoratus occur the most significant change and to explore the toxic effects of benzo[a]pyrene on S.marmoratus in depth from the aspects of gene,protein expression and enzyme activity on this basis.These study may provide a certain degree of scientific basis in monitoring and prevention of the control of PAHs as represented by benzopyrene in the marine environment.This article is divided into 4 parts:The first part is the preface.This part summarizes the biological characteristics and research status of the S.marmoratus,including its morphological characteristics and habits,growth and reproduction,and research progress and introduces the the overview of PAHs such as benzopyrene,their hazards and their toxicity to aquatic organisms,and the overview of research on cytochrome P450,including its composition,distribution,typing and nomenclature,and catalysis.induction and inhibition mechanisms.And it introduces the research progress of fish CYP1 A,including its induction and inhibition mechanisms,detection methods.The fish CYP1A1 gene,protein expression,EROD activity and five antioxidant defense system enzymes such as SOD induced by pollutants are also explained.The last discusses the purpose and significance of this study.The second part of this paper is the experimental materials and methods.This chapter mainly describes the experimental fish used in this experiment,primer,the main reagents and instruments used in the experiment,and the specific toxicity test methods,including concentration gradient experiments,time gradient experiments,the sample processing methods,the main solutions used in the experiment,experimental data processing methods and main software.The third part is the toxic effects of benzo[a]pyrene on S.marmoratus.Semi-quantitative RT-PCR was used to detect the effect of benzopyrene on CYP1A1 m RNA on the liver of S.marmoratus.Western blot was used to detect the effect of benzopyrene on the expression of hepatic protein in S.marmoratus.What's more,this paper studied the effects of EROD activity and the antioxidant defense system enzymes such as SOD,GPX,GSH,MDA and GST induced by Ba P.The results showed that the effect of CYP1A1 gene expression,protein expression and enzymatic activity of the S.marmoratus liver was induced by Ba P.That shows a certain concentration effect and time effect relationship.The fourth part is the conclusions and outlook of this experiment.By analyzing the toxicological effects of genetic,protein expression,and the enzymatic activity in the experiment on the toxic effects of the S.marmoratus,a comprehensive and systematic review of the indicators' role in the contamination of PAHs was conducted and indicators and their differences were analyzed.In future research,polycyclic aromatic hydrocarbons pollution are expected to provide guidance for the monitoring,prevention and control of marine organic pollution.
