Objective:With the development of society,faster pace of life,and the more intense competition,people are under great pressure.With the cost of overdraft health,eating irregular,and diabetes had become one of the biggest issues that faced by people today.The long course of these diseases requires long-term medication.Therefore,it is very urgent to study the hypoglycemic health foods.This product has the functions of lowering blood glucose,improving glucose tolerance and immunity.This prescription comes from the experience of Professor Zhang Tingmo,consisting of Mulberry Leaves,Puerariae Lobatae Radix,Ginseng Radix Et Rhizoma,Dioscoreae Rhizoma,Lilii Bulbus,and Eckloniae Thallus.The purpose of this study is to design appropriate preparation process,select the best extraction process,conduct systematic quality standard research,establish a suitable quality standard system,evaluate the in vitro and in vivo activities to provide scientific and effective basis for the later research and application of this product.Methods:Analyze the active ingredients of the prescription and the application status of various medicinal materials,combine the product efficacy and dosage form,designing a reasonable preparation process.Single factor test and orthogonal test were used to determine the best extraction process for Mulberry Leaves and Pueraria Lobatae Radix.Study the quality standardsof the product,including the inspection,identification,content determination,sensory evaluation and so on.Thin-layer chromatography?TLC?was used to identify each medicinal material,the total polysaccharide content was determined by UV-Vis spectrophotometry?UV?,the content of puerarin was determined by high performance liquid chromatography?HPLC?and establish the content limit of active ingredients.In the preliminary activity study,?-glucosidase method was used to determine the inhibitory activity of total polysaccharides on?-glucosidase,4-nitrophenol-?-glucopyranoside?PNPG?as a substrate and acarbose as a positive control.Established a mouse model of diabetes to further study the hypoglycemic effect of Sangge hypoglycemic powder.Results:1.The technological design of Sangge hypoglycemic powder:extract the active ingredients of Mulberry Leaves and Pueraria Lobatae Radix,Ginseng Radix Et Rhizoma,Dioscoreae Rhizoma,Lilii Bulbus,and Eckloniae Thallus pulverized into fine powder after pretreatment respectively,and mixed according to the prescribed dose by equal increment method.Orthogonal test results showed that the optimum extraction process of mulberry leaves was 50 times water extraction at 80°C for 3 times.Each extraction time was1.0 h.The polysaccharide yield and dry paste rate could reach 3.86%and 14.26%,respectively.The optimal extraction process was 12 times of 70%ethanol,80?water baths for 2 times,and each time was 1.5 h.The yields of isoflavones from Pueraria lobata diax and dry extract yields were 26.3%and 33.8%,respectively.2.The establishment of quality standards:Characters,inspections are in line with the regulations,indicating that the process is feasible.The TLC identification of Mulberry Leaves,Puerariae Lobatae Radix,Ginseng Radix Et Rhizoma is better specificity and can be included in the quality standard.The content determination results show that the linear relationship,reproducibility and stability of total polysaccharide and puerarin are good.The specificity of puerarin is good.The total polysaccharide content per bag should not be less than 2.3g;Pueraria isoflavone content?based on puerarin?per bag should not be less than0.7g.3.Determination of in vitro activity:The concentration-inhibitory effect of Sangge hypoglycemic powder on?-glucosidase is:y=0.1435ln?x?+0.7412?R2=0.9876?.The half-inhibitory concentration(IC50)value of Acarbose was 0.0648 g/L,the total polysaccharide of SangGe powder was 0.1862 g/L,the mulberry leaf polysaccharide was 0.3798g/L.Sangge hypoglycemic powder showed a good activity inhibitory effect on?-glucosidase,The activity of the enzyme was well inhibited,and its activity was significantly higher than that of mulberry leaf polysaccharide?P<0.01?.4.In vivo pharmacodynamic studies:The effect on body weight of alloxan-induced diabetic mice is not significant,but it can effectively reduce the water intake of sick mice.The medium-dose group?3.9 mg/kg?and high-dose group?7.8 mg/kg?both significantly improved fasting blood glucose?P<0.05?,and the blood glucose reduction rates were 33.13%and 27.77%,respectively.The effect was related to Yuquan Pills.There was no significant difference in the control group?decrease rate was 33.77%?.The medium and high dose groups also significantly increased the glucose tolerance in mice?P<0.01?.Conclusion:The experimental results show that the preparation technology of Sangge hypoglycemic powder is feasible,the quality is controllable.It has a good?-glucosidase inhibitory activity,it can effectively reduce blood glucose,and has a good auxiliary hypoglycemic effect.
