Diabetes is harmful to human health. At present the main treatment to control blood sugar is injected insulin and using hypoglycemic drugs. Most of the market hypoglycemic drugs are mainly designed to lower the blood sugar through chemical synthesis, but it has great toxic and side effects. Natural medicine is welcomed by diabetic patients because of less side effects and less toxicity. Recently, it has become a hot research to find the components with the hypoglycemic effect from natural products. Okra is a high nutritive vegetables.According to research report, okra has good hypoglycemic activity. This paper focuses on the extraction of okra polysaccharide and research its hypoglycemic activity. On the basis of the above, an okra lemon drink is developed. The main methods and results are as followed:First, we analyzed the basic components of okra. As a result, the total soluble sugar content of okra was 30.88%, protein content 16.23%, fat content 9.25%, reducing sugar 1.97% ,ash content 7.3%, and the content of cellulose 31.22%.The optimal extraction conditions of hot water extraction was discussed through single factor test and orthogonal test. The best condition is extraction temperature of 70 ?,extraction time 2 h, ratio of material to liquid 1:50, extracted 3 times. The extraction rate of okra polysaccharides was as high as 29.82% under the best condition.We established a mode of enzyme inhibitor in vitro to research the inhibitory effect of okra polysaccharide on a-glucosidase. In this reaction system, PNPG was used as a substrate, acarbose as the positive control. The contrast experiment of different purity of okra polysaccharides at the same concentration (2 mg/mL) shows the higher the purity of okra polysaccharide is, the stronger the inhibition rate on a-glycosidase enzyme is. The crude polysaccharide, non protein polysaccharide and purified component R1 inhibition rate of a-glucosidase were 38.28%, 51.85%, 62.93%, to verify the okra polysaccharide can reduce the blood glucose. We further analyzed the inhibitory activity on a-glucosidase of the three components, which were purified by DEAE-Sephadex-A25 chromatography column. The inhibition rate of R1, R2, R3 were 62.93%, 61.58%, 64.43%, without much difference. At last, comparing the hypoglycemic activity of okra polysaccharide under the different concentration, we found that the inhibitor on a-glucosidase activity increases along with the increase of okra polysaccharide, which showed a relation to doze. The IC50 was 2.393 mg/mL, which showed the okra polysaccharide had great inhibitory effect on a-glucosidase.STZ-induced hypoglycemic mouse model was used to study the hypoglycemic function of okra polysaccharide in vivo. The biochemical indexes of diabetic mice were detected after giving a gavage of non protein okra polysaccharide. The results showed that the okra polysaccharide had a significant effect on control blood glucose. It can not only reduce the content of TC, TG, LDL-C in the serum of mice,but also increase the content of HDL-C. It could effectively alleviate the phenomenon of lipid metabolism disorder caused by high blood glucose.Through the single factor and orthogonal experiment,we obtained the best formula of okra lemon drink: okra extract added 20%, lemon juice 12%, xylitol 9%. Adding 0.04%CMC-Na can maintain the stability of the beverage perfectly,under this condition, the beverage color is pale green, taste fresh and delicate, sweet and delicious. The beverage has okra lubrication taste and unique aroma of lemon.
