In this study,Lycium ruthenicum Murrr of Qinghai was used as raw material to extract anthocyanin.The microcapsule of anthocyanin of Lycium ruthenicum Murrr as core material was produced using beta-glucan ester as wall material.The technology of anthocyanin hydrophobically modified and the microencapsulation preparation have been studied.The stability mechanism,target release characteristics and functional evaluation of anthocyanins with microencapsulation were systematically studied.The main results of this paper are as follows.1.Using acetic acid as modifier,the anthocyanins of Lycium ruthenicum Murrr were modified by hydrophobic treatment.The optimum modification conditions of anthocyanin of Lycium ruthenicum Murrr were optimized by single factor and orthogonal test as follows.The addition of anthocyanin is 0.89%(m:v)in ethanol solution with a concentration of 50%,and the volume ratio of pyridine,acetic acid and ethanol solution(50%)is 3:7:450(v:v:v).The reaction time of modification is 4 h under 50 ?.Under these conditions,the solubility of anthocyanin is about 27.50% in the aqueous phase system,which decreases by 36.25% than that of unmodified anthocyanin solution.2.The highland barley ?-glucan was modified by 2-octenyl succinic anhydride as modifier.The optimum modification conditions of ?-glucan were optimized by single factor and orthogonal test as follows.Modification time was 5 h under 45 ?.The amount of addition of ?-glucan was 4.5 mg /mL and the addition ratio of 2-OSA was 0.5%(v:v).The C-O-C,C=O and a new double bond was successfully introduced into ?-glucan.3.The ?-glucan ester of highland barley was used as wall material of microcapsule,and the anthocyanin of Lycium ruthenicum Murrr was used as the core material.The encapsulation of anthocyanin of Lycium ruthenicum Murrr was prepared in aqueous phase system.The optimum preparation conditions of microcapsule solution were optimized by single factor and orthogonal test as follows.The concentration of highland barley ?-glucan ester was 2.5 mg/mL.Using agitator with input power of 5.0 W,the stirring time was 2 h under 45 ? and pH 3.0.The encapsulation rate of anthocyanin microencapsulated in aqueous system was about 46%.4.The effect of environmental factors(temperature,natural light,ultraviolet light and freeze-thaw cycle)on the stability of anthocyanin was studied by using the control variable method.The results showed that the thermal degradation of anthocyanin microcapsules was the first-order kinetics model.5.The retention rate of anthocyanin in aqueous system was taken as the index.The stability and targeted release characteristics of anthocyanin microcapsules were studied by simulating gastrointestinal fluid in vivo and in vitro.In vitro simulation of gastrointestinal fluid stability test,anthocyanin microcapsules were more stable in simulated gastric fluid than in the intestinal fluid,and the retention rates of anthocyanin microcapsules were increased by 18.4% and 24.7% in simulated gastric fluid and simulated intestinal fluid compared to the unencapsulated anthocyanin solution.Anthocyanin release rate was 7.5% and 20.7% for anthocyanin microcapsule solution and anthocyanin solution after 90 min in the simulated gastric fluid.Anthocyanins were released slowly in simulated intestinal fluid,and release rate respectively were 21.9% and 27.2% for anthocyanin microcapsule solution and anthocyanin solution after 240 min.The stability of anthocyanin microcapsule in vivo could be increased to ensure the absorption and utilization of anthocyanin in human body.6.The acute toxicity test showed that there was no toxicity of anthocyanin microcapsule solution of Lycium ruthenicum Murrr.In order to study the anti-fatigue effect of anthocyanin microcapsule solution of Lycium ruthenicum Murrr,The weight-bearing swimming test has been done,and content of blood lactate,liver glycogen and serum urea nitrogen have been detected.It was found that the appropriate solution was low dose level of 1.0 mg/mL for good anti-fatigue effect.The swimming time was increased by 89.89%,and the clearance rate of blood lactate was 22.82%,and the content of glycogen in liver was increased by 123.55%,and the level of serum urea nitrogen was decreased 23.12% compared to he blank control group.In the antioxidation test,it was found that the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in serum and liver tissue were increased and the content of MDA and lipofuscin in tissues were decreased by microcapsule solution of anthocyanidin of Lycium ruthenicum Murrr.The high dose concentration(8 mg/mL)of anthocyanin microcapsule solution of Lycium ruthenicum Murrr was the best for antioxidation.
