Lycium ruthenicum Murr., belonging to natural anthocyanins pigment resourcewith abundant amaranthine pigment in its ripe berry, is a kind of halo-tolerant anddrought-resistant Solanaceae lyceum in northwest of China. In order to stimulateconverting our Western plant resources into economical ones and provide a feasibleapproach for soline-alkali soil industry developing, the present research has verifiedthe economic and pharmaceutical usefulness through the study of the extracting andrefining technology and biological activity of anthocyanins and Polysaccharides fromLycium ruthenicum vinasse. This research relatively offers a theoretical reference onthe development and use of Lycium ruthenicum Murr.In order to study the extracting and refining technology of anthocyanins inLycium ruthenicum, we get the best extracting condition by orthogonal experimentand Response Surface Methodology. It is ph1.0distilled water used40times volumethan that of dried fruit,0.8%pectase,50℃,60minitues; The X-5resin is used torefine the pigment, the best adsorption condition is1g/L pigment solution, pH3.0,3mL/min, diament VS height:1:15, which adsorption rate is95.43%; The95%ethanol is used as eluting solvent, pH2.0,4mL/min,3BV eluting solvent volume arethe best desorption conditions, the anthocyanins produced by this method is purplepower, and its colour value is36.5. The technique is simple and convenient, furthermore, it has good repeatability.According to the same way, we get the best extracting condition forpolysaccharides is distilled water is used as impregmengt,90℃,60minitues,repeating for5times, then the content of polysaccharides is maximum with42.75%;The AB-8resin is used to refine the polysaccharides, the best adsorption condition is1g/L polysaccharides solution, pH9.0,1mL/min, diament VS height:1:15; The pH9.0,1mL/min,0.25%eluting solvent,5BV eluting solvent volume are the bestdesorption conditions, the polysaccharides produced by this method is white power,its content is67.13%.The study on antioxidant activity in vitro experiment shows that bothanthocyanins and Polysaccharides have the ability. The IC50in hydroxy radical,DPPH radical, superoxide anion radical and lipidosome overoxidated byanthocyanins Polysaccharide are3.43,2.47,3.51and4.74mg/L. The IC50in hydroxyradical, DPPH radical, superoxide anion radical and lipidosome overoxidated byPolysaccharides are2.01,2.81,2.42and2.95mg/L. The main ingredient of anthocyanins from Lycium ruthenicum vinasse is Petunidin-5-O-glucoside,accounting for42.91%through HPLC methology.The results of hypolipemic and anti-atherosclerosis experiments on Polysacch-arides from Lycium ruthenicum vinasse show that the the polysaccharides candecrease TC, TG and LDL-C and increase HDL-C in the hyperlipoidemia mice, itcan also increase the activity of T-AOC, GSH-Px, SOD in mice’ livers and decreaseHDL-C. The pathological section shows that the amounts of AI in different dosagesof Polysaccharide groups are much lower than in the reference ones and thepathologic changes in aorta, heart and liver are much less than in hyperlipoidemiagroups. Amount the test, the effects of standard dosage of Polysaccharide groups,which are close to simvastatin groups,are much better than in higher and lowerdosage ones.
