With the development of aerospace technology,the effects of mechanobiology on creature in extreme mechanical environment—high acceleration are becoming an issue of concern.Studies have shown that high acceleration also has effects on organs,tissues,cells and molecules.For the high acceleration environment simulation,aerospace sectors have special equipments for high acceleration loading,such as the vertical drop tower and manned centrifuge,etc.But for the general laboratory,the study on mechanical and biological effects of organisms under high acceleration environment is short of conditions.In the laboratory,the function of the ordinary centrifuge is mainly used for centrifugal separation and sample preparation.Centrifuge can be used for cell loading is very scarce,especially for adherent cell loading is more less.The existing centrifuge has been unable to meet the requirements of the present experiment.According to current ordinary centrifuge and cell culture environment,this paper design a new type of centrifugal device for adherent cells loading,which has the characteristics of small volume,low noise and high safety performance.In this paper,a centrifugal machine for high acceleration loading on adherent cells is developed,and the angle adjustable rotor is designed.First of all,this paper analyzes the structure and working principle of high acceleration loading device at domestic and foreign,and summarizes their advantages and disadvantages in the application.Combining the experiment requirements in this subject,the design indexes are determined that the centrifugal machine can realize 0 to 40 g constant acceleration loading and 0 to 0.8g/s variable acceleration loading,and the design scheme of the centrifugal loading device is made.Then,a3 D model of the body structure is established,and the rotating electric machine and other parts are selected.For the rotor in centrifugal machine for high acceleration loading,its characteristic is that culture plate and culture bottle can be loaded directly.A 3D model is established by using SolidWorks first.Then the finite element analysis is carried out on the rotor model using finite element analysis software ANSYS.After determining the rotor structure meets the requirements of strength and safety,the rotor is manufactured.When using the rotor,culture plates and culture bottles should be filled with nutrient fluid before clamping on them,to prevent the existence of bubbles between the adherent cells and nutrient fluid during loading and impact loading effect.Moreover,the culture plates must be sealed and clamped with the clamping plate and the culture bottle caps tighten,in case of nutrient fluid leakage.In addition,the culture plates and cultures sealed should be balanced,to ensure that the quality of rotor symmetry on both sides is evenly distributed.Furthermore,angle adjustable rotor can give constant acceleration and gradient loading according to different angles,and also can be used for animal centrifugal load.In the same,a 3D model is established by using SolidWorks first for the angle adjustable rotor to determine the rotors can meet the requirements on the function.And the finite element analysis of the angle adjustablerotor is made by using ANSYS Workbench,analyzing of stress and strain in different angles.Analysis results show that the structure of the rotor can meet the requirements of strength and safety.This subject combines the mechanical design and biomechanics,the centrifugal loading device designed in this paper can be used for the exploration of cell mechanobiology under high acceleration.Research of this new type of centrifugal loading device provides a convenient way for centrifugal loading on cells,also provides a new way for centrifugal loading researches in colleges and universities,companies and scientific research units.
