Spatial microgravity has always been a significant factor affecting organisms in space environment.Monitoring of spatial microgravity damage is of great significance to the research of space biology and the development of spaceflight.At present,the technology of space microgravity damage monitoring is based on DNA monitoring,protein monitoring and observation of nematode life status.These monitoring techniques have disadvantages that include difficult operations and strong professional knowledge,meanwhile,the equipment is sophisticated,complex and bulky.In view of the above problems,this paper presents a monitoring method of space microgravity damage,and designs a monitoring system of space microgravity damage.The method takes C.elegans as a model organism,and the number of luminescent sites,the fluorescence intensity and the aspect ratio are taken as the evaluation.indexes for monitoring microgravity damage.Microfluidic chips are used to select,cultivate and fix nematodes.A miniaturized microscopic imaging module was designed to capture nematode fluorescence and bright field images.Through the nematode fluorescence and bright field images processing,we obtained three evaluation index of spatial microgravity damage.Therefore,the system has these advantages of small size,simple structure,low price and simple operation.The working principle of this system is to cultivate and immobilize Caenorhabditis elegans in the microfluidic chip and acquire the fluorescence and bright field images in real time,and obtain the evaluation index of spatial microgravity damage monitoring by the fluorescence and bright field image processing of Caenorhabditis elegans.The system consists of microfluidic chip,microscopic imaging module,excitation light source module,bright field light source module,micro pump and PC.Through the mechanical structure design,it is integrated into a complete set of space microgravity damage monitoring system.In this paper,microscopic imaging experiments were carried out to confirm the good imaging quality of the microscopy imaging module.In this paper,simulated microgravity equipment was used to deal with C.elegans,and comparative experiments between this system monitoring and microscopic monitoring were carried out to verify that the monitoring method is feasible and the monitoring system is accurate,stable and reliable.
