As a disadvantageous result of antibiotics on feed, new substitutable feed additives are needed extremely urgent. Polysaccharides from the Submerged Fermentation extract of H.caput-medusae (Bun.:Fr) Pens (HFP) is one of nature and non-toxic Fungi polysaccharides. Many studies show that HFP have functions of antitumor, antivirus and antimicrobial.240healthy Avian broiler chickens at1day of age with similar growth performance were randomly divided into4groups with3replicates of20chickens in each group feed by0,0.1%,0.3%,0.5%HFP (mass ratio) added in non-antibiotics corn-soybean basal diet in42days trial period. Confirmatory analyses of variations in growth performance, intestinal pH, intestinal flora amount, caecum short chain fatty acids (SCFA) and microbiome composition showed the influence of different levels of HFP in meal diet on broilers intestinal microflora. The achieved results provided important theoretical base for the industrial production of HFP.The results as followed:1.To a certain extent, HFP could promoted the production performance of broilers.0.1%HFP could have no significant effect on the average daily gain, the average day intake and the feed conversion rate.0.3%HFP could decreased the average daily gain, the average day intake and the feed conversion rate.In the early stage of experiment, the feed conversion rate of0.3%HFP was significantly lower than that in blank group (P<0.05). In the later stage of experiment, the feed conversion rate of0.3%HFP was no significant than that in blank group (P>0.05).To a certain extent,0.5%HFP could promoted the average daily gain and decrease the feed conversion rate.2.0.1%HFP group could extremely significant decreased Ileum pH value of14days broilers (P<0.01).0.3%HFP group could significantly decreased Caecum pH value of21days and Ileum pH value of28days broilers (P<0.05).0.5%HFP group could significantly decreased Caecum pH value of21days and Ileum pH value of28days broilers (P<0.05), and extremely significant decreased Caecum pH value of42days and Ileum pH value of14days broilers(P<0.01).0.1%,0.3%.0.5%HFP groups significantly increased the amounts of Bifidobacterium and Lactobacillus in Ileum and Caecum (P<0.05),decreased the amounts of E.coil, Salmonella and Enterococcus.Expecially,0.5%HFP group had the best effect.3. Feeding the adding HFP in daily diet could increased the concentration of acetic acid, propionic acid and butyric acid. In28age, compared with the blank group, the concentration of cecum acetic acid of0.1%HFP reached significant difference (P<0.05); the concentration of the cecum propionic acid of0.1%,0.5%HFP group reached significant difference (P<0.05); the concentration of cecum butyric acid of0.1%HFP group reached extremely significant difference (P<0.01). In42days of age, compared with the blank group, the concentration cecum propionic acid of0.1%HFP reached significant difference (P<0.05), the concentration of cecum propionic acid0.5%HFP group reached extremely significant difference (P<0.01); the concentration of cecum butyric acid0.1%HFP reached significant difference (P<0.05), the concentration of cecum butyric acid of0.5%HFP reached extremely significant difference (P<0.01).4.Through the ERIC-PCR reaction system for various factors in the single factor and orthogonal tests, amplification procedures and amplification system for the analysis of ERIC-PCR was established. The optimized ERIC-PCR stable reaction system(25?L):10×buffer2.5?L, dNTPs0.2mmol/L, Mg2+2.5mmol/L, DNA template52.5ng, primer14.5pmol. Taq DNA polymerase2.5U. The optimized amplification program was:?95?for7min?94?for1min,52?for1min,65?for1min,30cycles?65?for10min; hold on4?.5.According to the reaction of the sample that the cecum DNA fingerprint found ERIC-PCR of the blank group and HFP group (0.1%,0.3%,0.5%) of14,21,28,42days of age broilers,there are differences from them in the cecum and ileum intestinal floar structural. Specific strip significantly were more than blank group.Feeding HFP resulted in the flora structure were more abundant.Broilers were healther and more benefit at adjusting the intestinal environment balance.
