ObjectiveChinese Crude Drug GAP,the English abbreviation for the good agricultural practice for traditional Chinese drugs,was published by CFDA in April 2002 to control the effective factors ofquality of Chinese Crude Drug and standardize every step throughout all production processes,to reach the purpose of'authentic,superior,stable and controllable"for Chinese Crude Drug.For guaranteeing the smooth implementation of GAP(trial implementation),CFDA formulated and published GAP certification management method(trial implementation)and GAP compliance standard(trial implementation)in September 2003.And since November 1st,2003,CFDA started accepting application of GAP certification submitted by Chinese medicine enterprises.SPATHOLOBICAULIS,a Chinese medicine commonly used for blood-activating and stasis-dissolving,which is the dried rattan of Spatholobus suberectus Dunn,specified by Chinese Pharmacopoeia(first part of 2010 Edition).It is an important raw material of patent medicines "zhengtian pill " and "san-jiu strong bone joints pills",which has a high market consumption manutactured by China resources Sanjiu medical&pharmaceutical CO.,LTD.Due to the plant' slong growth periodandover-felling in recent years,the wild resource of Spatholobus suberectus Dunn has begun exhausted,therefore,developingits standardized planting and establishing GAP base becomes the way of solving the resource problem.A Spatholobus suberectus Dunn GAP base in Pingyuan county,Guangdong province,which was belonged to China resources Sanjiu medical&pharmaceutical CO.,LTD,supported this thesis to carry out investigation of the technical regulations for Spatholobus suberectus Dunn production to meet requirements for GAP authentication of Spatholobus suberectus Dunn,has and to provide technical support for the GAP certification application of subsequent related enterprises.Methods and Results1.Experimental study1.1 Study on Spatholobus suberectus Dunn seedling breedingThree cutting processing methods were set,namely direct cutting,black plastic film protected cutting,and seedling bag protected cutting;and hormone soak shoot cutting and hormone root filling were also set as different hormone treatment ways,and 3 hormone concentrations(0 mg/L?300 mg/L?600 mg/L)were used to conduct the single factor experiment to optimize Spatholobus suberectus Dunn seedling breeding.The results showed that rooting percentage could reach over 80%under the optimal seedling technique(seedling bag protected cutting)and the use of 300 mg/L ABT1 by root filling.1.2 Quality classification of Caulis Spatholobus suberectus Dunn seedlingsThe Seedling height,branch number and stem diameter of 500 local bredSpatholobus suberectus Dunn seedlings were measured,and calculated to obtain the average value and standard deviation,which were then used to classify the seedling quality.The results showed that the 3 indexes of first-class seedlings were no less than 7.4cm,3 and 3.49 mm,respectively;theindexes of second-class seedling werebetween 1.6 cm and 7.3 cm,2 to 3,and 2.38 mm to 3.48 mm,separately;while the below standard seedling were not higher than 1.5 cm,1 and 2.37 mm respectively.1.3 Study of soil testing and formulated fertilization for Spatholobus suberectus Dunn plantingThe soil fertility of Caulis Spatholobus suberectus Dunn planting base in Pingyuan county of Guangdong province was measured and the soil fertility status of breeding bases and field industrialization base were also diagnosed.The results indicated that soil pH and exchangeable calcium and magnesium contents were unbalanced and the contents of organic matter and Nitrogen and Phosphorus which belong to major elements,were slightly low.A fertilization scheme was made based on the soil fertility conditions and referred to technical specification of balanced fertilization by soil testing as followed:(1)Using soil conditioners such as bentonite,gypsum and hydrated lime to adjust the soil pH;(2)Using micronutrient fertilizer solution to treat cutting slips in the process of growing seedlings,followed by urea fertilized with 5 kg/667m~2;(3)Using rotten fertilizer as base fertilizer in transplanting engraftment,its dosage was 10 kg for each plant.(4)Based on the fertilization of compound fertilizer in transplant,amrmonium dihydrogen phosphate(15kg/667m~2)was used to strengthen the supplement of N and P,and the appropriate amount ofhydrated lime and magnesium carbonate were used to supplement soil microelements.1.4 Investigationof Spatholobus suberectus Dunnharvesting season based on herb qualityThe contents of effective elements of Spatholobus suberectus Dunn samples were measured monthly,it is shownthat catechin and epicatechin were higher from January to June,the highest among them was harvested in March,which reached 0.8866 mg/g;while the highest epicatechin content was 2.0932 mg/g that was obtain from the samples collected in January,the total content of catechins was higher in January and February.Therefore,the best harvesting season of Spatholobus suberectus Dunn in Pingyuan county of Guangdong province was winter.2 Formulation of related operation procedures of other items2.1 Operation specification of medicinal materials quality inspection of Spatholobus suberectus Dunn,including the content of internal control quality standards of Spatholobus suberectus Dunn medicine enterprise,was formulated based on the results of previous studies.2.2 According to literature review,6 operation procedures wereformulated in the thesis,namely irrigation and drainage operation procedures of Spatholobus suberectus Dunn production,field management operation procedures of Spatholobus suberectus Dunn production,pests and diseases prevention plans of Spatholobus suberectus Dunn,medicinal materials sampling operation procedures of Spatholobus suberectus Dunn,packaging operation procedures of Spatholobus suberectus Dunn and storage and maintenance operation procedures of Spatholobus suberectus Dunn.conclusionBased on experimental researches and literature researches,this paper has improved and supplemented many technology regulations of Spatholobus suberectus Dunn production that were required to authentication by GAP(trial implementation);and has drafted the SOP,which has initially constructed the GAP authentication system of Spatholobus suberectus Dunn andprovided technical basis for the GAP certification applycation of subsequent related enterprises.The GAP technical documents and experimental initial data that were generated in this paper were incorporated into the GAP tracing database of Spatholobus suberectus Dunn,which was constructed by the research group to establish the medicinal materials quality tracking system of Spatholobus suberectus Dunn production.
