Effects Of Complex Chinese Crude Drug "Zhukangsan" On Antioxidantion In Serum And Tissues Of Weaned Piglets Artificially Prevented By PRRSV

The aim of this study was to investigate the effects of different dosage of complex Chinese crude drug "Zhukangsan" on the levels of antioxidantion in serum and tissues of weaned piglets artificially prevented by PRRSV, and in order to reveal the prophylactic effect of complex Chinese crude drug "Zhukangsan" on PRRSV, then provide scientific basis for pig production. Forty piglets of 35-day-old were divided randomly into 4 groups treated with virus and negative group (I), positive group (II),0.5%(Ⅲ), and 1.0%(IV). Group Ⅲ and Ⅳ were given drug (Zhukangsan) at 0.5% and 1.0% in the forage at 41 to 44-day-old, respectively, then all groups except group I were inoculated PRRSV SCM at 41-day-old. At 40,45,50,55 and 60 days of age during the experiment, the blood was sampled to separate serum and measured the serum antioxidant indexs from each experimental group of four piglets. And at 50, 60 days of age during the experiment, the liver, lung, spleen and kidney were collected for tissue antioxidant indexs from each experimental group of four piglets.Results:(1) During the whole experiment period, compared with the negative group and the positive group, the two groups of traditional Chinese medicine could improve markedly (P<0.01 or 0.05) the levels of SOD in serum of piglets challenged by PRRSV except 40 day; Except liver and lungs on 50 day, the values of SOD in tissue of piglets challenged by PRRSV of the group of 1.0% traditional Chinese medicine markedly higher (P<0.01) than those of negative group and positive group; When the 60 day, the liver, spleen and kidney SOD of the 0.5% group were superior (P<0.01) than positive group.(2) Compared with the negative group and the positive group, the levels of GSH-Px in serum of 0.5% and 1.0% traditional Chinese medicine were significantly greater (P<0.01) except 40 day and the 0.5% group on 45 day and 60 day; Except pulmonary of the 0.5% group and spleen on 50 day, The values of the tissues GSH-Px in the 0.5% and 1.0% traditional Chinese medicine group were higher (P<0.01) than those of the positive group.(3) Compared with the negative group and the positive group, the two groups of traditional Chinese medicine could improve markedly (P<0.01) the levels of CAT in serum of piglets challenged by PRRSV except 40 day and the 0.5% group on 45 day; Except lungs of the negative group on 60 day, the values of CAT in tissue of 1.0% traditional Chinese medicine were higher (P<0.01 or 0.05) than those of negative group and positive group; Except lungs on 50 day, the levels of CAT in tissues of 0.5% group were superior (P<0.01) than positive group.(4) Except 40 day and the 0.5% group on 45 day, the levels of T-AOC in serum of piglets challenged by PRRSV of the groups of 0.5% and 1.0% traditional Chinese medicine were significantly higher (P<0.01) than those of the negative group and positive group; The values of T-AOC in tissues of 1.0% traditional Chinese medicine were higher (P<0.01) than those of negative group and positive group, the levels of T-AOC in tissues of 0.5% group were superior (P<001) than positive group.(5) The contents of the serum MDA in the 1.0% traditional Chinese medicine group were significantly lower (P<0.01) than those of the negative group and positive group except 40 day and the 0.5% group on 45 day; except kidney, lung and spleen on 60 day of the negative group, the contents of the tissues MDA in the 0.5%and 1.0% traditional Chinese medicine group were significantly lower (P<0.01) than those of the negative group and positive group, the contents of MDA in tissues of 0.5% group were lower (P<0.01) than positive group.(6) During the whole experiment period, compared with the negative group and positive group, the levels of the serum inhibition of OH capacity in the 0.5% and 1.0% traditional Chinese medicine groups were significantly superior (P<0.01) except 40 day; The levels of the tissues inhibition of OH capacity in the 0.5% and 1.0% traditional Chinese medicine groups were significantly higher (P<0.01 or 0.05) than those of the positive group except liver on 50 day and kidney on 50 day of the negative group.In conclution,0.5% and 1.0% complex Chinese crude drug "Zhukangsan" in the forage could improve the levels of the activity of the antioxidant enzyme of serum and tissues of piglets artificially prevented by PRRSV, act to scavenge free radicals and improve body function, thereby build up resistance of piglets to PRRSV. The optimum traditional Chinese medicine add dosage was found to be 1.0%.
