Fusarium Wilt is a common disease in cotton which performs seriously at present in the sea island cotton(Gossypium barbadense). Its occurrence and development will directly affects yield and quality of cotton.This studywas conducted to Gossypium barbadense Fusarium Wilt disease resistance genetic analys is and resistant germplasm resources evaluat ion, including an F2:4 population constructed by the resistant parent Pimas-7 and the susceptible parent parent 5917, an natural populat ion constructed by 143 sea island cotton germplasms and an RIL population constructed by the susceptible parent parent Xinhai 14 and resistant parent 06-146. Combining the Gossypium barbadense Fusarium Wilt disease resistance related the research of gene expression and the selection of disease resistance marking to award resistance maker.First, The research realized better Fusarium Wilt resistance groups through the investigation and evaluation to the field disease index P1,RI L line P3 and the germplasm of Gossypium barbadense P2. Secondly,by literature and using NCBI website to get a sequence In order to find the label related to Sea Island cotton Fusarium wilt and providing benef icial reference for the Sea Island cotton disease resistance molecular assisted breeding. Integrated the study,the conclusions were as follows:1.The research investigated the phenotypic traits in the field of Fusarium Wilt resistance in three different genetic populations, The results showed that :(1) through making statistics in different levels for Fusarium Wilt resistance of sea island cotton, it was found that all of three populations performed positive segregation distribut ion both in the disease levels and the disease index, and there were super parent materials in both P1 and P3 of two resistance traits;(2) through analysis of variance it was found that there was significant differences among three different populations both in the disease levels and the disease index, P2 with highest value in two resistance traits but least in CV and P3 with least value in two resistance traits but highest in CV;(3) through heritability estim ation it was concluded that the hB2 of f usarium wilt resistance in sea island cotton was about 70%, belong to the medium her itability;(4) through clustering these 143 sea island cotton germplasms could be divided into four groups according to the two resistance traits and the group included the variet ies selected in Xinjiang performed best in f usarium wilt resistance than others.2.Using the primer design from the 4 EST sequence associated with stress,defense-related,cell wall structure,growth,photosynthesis of paragraph. And by literature and using NCBI website to get a sequence from cotton MAP K genes associated with group D and related to disease resistance,and Sequencing Laboratory obtained sequence associated with Pathogenesis-related proteins, transcription factor,glutathione S-transferase,gossypol,flavonoids,glycosyltransferase,then, fluorescence quantitative primer on thhese sequence, using the designed primers for disease resistance related gene expression characteristics analysis under the condition of Fusarium wilt induced get Gossypium barbadense and upland cotton resistance materials and Infected material. The results showed that the CF,CFW3,CFW8,CFW10, CFW17 comp71372?comp60501?comp62329?comp65705?comp55896 and comp62810 were signif icantly expressed in the upland cotton and Sea Island cotton. And Distinguish resistant and susceptible island cotton material.3. The use of 6 pairs of markers with the resistance to Fusarium Wilt with linked to carry on the related analysis between disease stage and Agronomic Traits in an RIL populat ion constructed by the susceptible parent parent Xinhai 14 and resistant parent 06-146, then get the positioning mark of 014, 016, BNL1066 with Fusarium Wilt of cotton was significantly related to disease stage. And these 3 pairs can be used as effective genetic markers for the correlation analysis of Fusarium wilt resistance in Sea Island Cotton. Now we could verify these 3 markers in an F2:4 population constructed by the resistant parent Pimas-7 and the susceptible parent parent 5917, to identify the resistant and susceptible materials, then using a combination of markers of 014-R/016-R/BNL1066-R can be more effective to choose disease resistance materials of high resistance to Fusarium wilt.
