In this study, a survey on the Fusarium wilt resistance of 80 cotton germplasm materials was carried out in two years (2014 and 2015), Then, the two factors variance analysis and correlation analysis of the disease index data and output data were carried out for two years. At the same time, by referring to the literature and the transcription group sequencing, the resistance related genes or EST sequences were obtained. The induction of Fusarium wilt was tested by 06-146, XH14,10893,10895,10897,10796, after 40 hours of inoculation, the extraction of RNA from the embryonic axes was extracted. Using the extracted RNA to carry on the analysis of the gene expression characteristics of q-PCR, the results were obtained:1. The disease index, disease level, incidence, yield and other related data were calculated, according to the field investigation of two years and the relevant formula, the results were obtained:Double factor analysis between the disease index and the year, found that the disease refers to the difference between the years is very significant, the difference between the varieties of significant differences; Cluster analysis of all the materials,80 germplasm resources can be divided into 3 categories; Through the analysis of the variance between the average individual plant yield and the year, it can be concluded that the yield difference was not significant, but the difference was significant between the different resource materials and the yield; Correlation analysis between disease index and yield showed significant negative correlation between yield and disease index.2. According to the analysis of the data from the use of the transcription group sequencing, the results were obtained:In samples infected with Fusarium wilt, the resistance related genes were significantly expressed in the disease resistant materials, and the expression level of most of the genes was significantly higher than that of the susceptible samples. It include:Resistance response protein gene PR 1 and PR3, toxin related genes GSTF and GSTU, Defense mechanism related genes MRP and NPR, gossypol biosynthesis related gene ACLA, R gene ZAR1 and RPS2 et al. The functional clustering results of differentially expressed genes showed that the genes related to the sugar-based transfer enzymes and flavonoid biosynthesis genes were enriched in the samples of infected Fusarium wilt. The genes in these metabolic pathways are related to the disease resistance of cotton, and it can be used as a candidate gene for the study of disease resistance in cotton.3. By using the collected 11 genes to make fluorescent quantitative analysis, the difference of gene expression was analyzed, the results were obtained:CFW3, CFW10, comp62810, comp71372 genes were significantly different in the process of infection of Fusarium wilt. The difference is mainly reflected in the following:relative to the susceptible materials, resistant materials with the highest expression level of time than sense disease materials early expression; far higher than susceptible disease materials. It is speculated that these four genes may be associated with cotton resistance to Fusarium wilt.
