Pepper (Capsicum annuum L.) is the second largest vegetable crop in China, which plays an important role in the world. Because of it's obvious heterosis, pepper hybrid F1 seeds have been widely used to increase yields and improve the quality of peppers. But due to artificial selection pressure term, The genetic basis of conventional cultivars has gradually narrow, intraspecific hybridization has been unable to meet the genetic improvement. For planting innovation, Interspecific hybrid of pepper particularly with cultivar (cultivated varieties and wild species) interspecific hybridization has become the focus of the study. In order to enhance the genetic basis, obtaining the hybrid plant have advantage. The research is based on thick purple pepper C.annuum PBC1366 or wild species C. chacoense PI260426 was reciprocal crossed with five cultivated species, and using embryo rescue technique to cultured the hybrid embryos successfully obtaining two combinations of hybrid plants. After using morphological observations?fertile with stained pollens and EST-SSR molecular marker technique to identify hybrid plants, The results were summarized as follows:1. With manual pollination, an interspecific hybrid was obtained from the cross between thick purple pepper C.annuum PBC1366 as female parent (P1) and strong spicy pepper C.chinense PI439487 as male parent (P2).19 phenotypic traits were observed and compared among Pi, Fi and P2. The results showed that the F1 had heterosis significantly and majority of the other phenotypic traits were intermediate to that of either parent. The pollen stainability percentages of interspecific Fi, the nature of hybrid was confirmed by morphological observation and EST-SSR marker analysis.2. Hybrid plants from the combinaton of C.chacoense PI260426× C.chinense PI152225 was obtained through reciprocal interspecific hybridizations among the C.chacoense PI260426?C.chinense PI152225. And putative F1 plant obtained through embryo rescue technique, the C.chinense PI152225 as female and the C.chacoense PI260426 as male parent. After using morphological observations?fertile with stained pollens and EST-SSR molecular marker technique to identify hybrid plants.The obtainment of the interspecific hybrid plants afford a basis for genetic map construction, transfer of the fruit flavor gene, anthocyanins metabolic path way in pepper.
