In the need of crop variety examination and approval work in Anhui province,it must be built the culture collection of the planting zone in Anhui province,for the service of Authorized variety of disease resistance identification.wheat scab?wheat sharp eyespot?Bipolar maydis?corn basal stem rot are the important diseases on wheat and corn production in Anhui province,they also are the main objects to submit for approval breed disease resistance identification.The foundation work of the culture collection is to screen of optimize the storage condition of the above disease pathogens,it's of great significance.In this thesis,Based on the above four kinds of bacteria as the research object,it is mainly by mycelium growth rate,spore production quantity of bacteria and pathogenic force index as the basis,it through the base material,different storage temperature and different base material moisture content and different save the time of impact test,to find out the better pathogen for the combination of optimum storage conditions,It service for wheat,corn varieties identification and disease-resistant breeding of disease resistance.The main results are as follows:the study of lyophilization method for the base material moisture content After different vacuum freeze drying time of 4 kinds of base material(wheat,barley,corn,gram)to study the influence of water content,it has the relation between vacuum freeze drying time and moisture content,It is good for germs stored when the vacuum freeze drying time is 22 h-33 h,the solid medium content water content below 10%.optimization screening the strains of storage methods: the study of wheat,barley,maize,gram four kinds solid medium: it sets 0 h,11 h,33 h,55 h after the freeze drying time of processing,They are preserved in 4 ?,-20 ? and-80 ? temperature,The mycelium growth rate ?the amount of spore production and pathogenicity are determined at different storage period,.Results show that in the 4 kinds of solid medium,The viability of wheat scab?wheat sharp eyespot in the corn solid medium is the best.setting four medium moisture gradient conditions,preserving in-80 ? temperature preservation for 3 months,the mycelium growth rate of wheat Fusarium graminearum were 3.10 cm/d,The sporulation were 3.98 x 105/ mL,the disease indexes were 0.87.Wheat sharp eyespot's disease indexes are 0.48 on average.The viability of Bipolaria maydis?corn basal stem rot in the wheat solid medium is the best.Preserving in-80 ? temperature for 3 months,Bipolaria maydis the average of sporulation were 5.63 x 105/ mL,the disease indexes were 0.61.Corn basal stem rot spore production amount to an average were 2.18 x 10~7/ mL,the condition of field identification index were 0.76.Determine the storage time of strains storing,storaged in-80 ? temperature conditions,It setted up 12 months,15 months,18 months three kinds of treatment,and measured the mycelium growth rate of wheat Fusarium graminearum in corn culture medium were as follows: 2.316,2.414,2.265(cm/d).The average hyphae growth rate of corn basal stem rot in wheat medium as follows: 2.306,2.418,2.193(cm/d).With the storage time prolongingzhen,the epiphyte growth rate was slightly reduced.
