Study On Candidate Genes Of Small Intestine Development Of Weaned Piglets

This study is on the Anhui local breeds Anqing Six White pig compared with world-class lean-type pig Landrace of weaning for the 28-day-old piglets. Indicators of serum contents, intestine mucosal morphology, small intestine function and gene expression were all examined at the 33-day-old and 39-day-old piglets, in order to investigate the differences of small intestine development related index, related genes expression levels and correlation between gene expression and intestinal development of various indicators at weaning stress stage between chinese pig and world-class pig. The results could provide the basis for the follow-up study of the related candidate genes during the weaning stress period for piglets, and reference for the early weaned stress and the related studies of the Intestinal development of piglets. The results are as follows:1) The GH, GA, T3, T4, Cor, CD3 and T-AOC in the serum of Anqing Six White pigs were significantly higher than that of Landrace at the 33-day-old(P<0.05); the GH, GA and Cor, CD3 and Ig G in the serum of Anqing Six White pigs were significantly or extremely significantly lower than that of Landrace(P<0.05, 0.01), the T3 and T4 in the serum of Anqing Six White pigs were significantly or extremely significantly higher than that of Landrace at the 39-day-old(P<0.05, 0.01).Comparison of the same pig breeds, the GH, Ig G, Ig M, T-AOC and Ig A in the serum of Anqing Six White pigs and Landrace were extremely significantly improved(P<0.05), the GA and Cor in the serum of Anqing Six White pigs were significantly or extremely significantly decreased(P<0.05,0.01); the GA was significantly improved(P<0.05), the T4 was significantly decreased in the serum of Landrace(P<0.05).2) The small intestine villus height of Landrace were higher than that of Anqing Six White pigs at the 33-day-old and the 39-day-old. The small intestine villus height, crypt depth of ileum, villous height/crypt depth of duodenum and jejunum of Landrace were significantly or extremely significantly higher than that of Anqing Six White pigs at the 33-day-old(P<0.05, P<0.01).The villous height and crypt depth of duodenum and ileum of Landrace were significantly or extremely significantly higher than that of Anqing Six White pigs at the 39-day-old(P<0.05,P<0.01). It showed the change trend of the small intestine villus height of Anqing Six White pigs and Landrace at the 33-day-old and 39-day-old were duodenum was the highest, followed by jejunum, ileum was the lowest,which reached a significant level.3) There were variety differences of small intestine disaccharide enzyme and mucosal morphology between Anqing six white pigs and Landraces at the 33-day-old and 39-day-old, which reached a significant or extremely significant level(P<0.05, 0.01). Comparison of the same Pig breeds, at the 39-day-old, the maltose activity, Lactase activity of duodenum,jejunum mucosa,the Na+, K+-ATP Activity of duodenum, jejunum mucosa of Anqing Six White pigs were significantly or extremely significantly decreased at the 39-day-old(P<0.05, 0.01); Ca2+,Mg2+-ATP Activity of jejunum mucosa,Sucrose Enzyme Activity of duodenum, ileum mucosa of Anqing Six White pigs were significantly or extremely significantly improved(P<0.05, 0.01).The change tend of disaccharide enzyme and ATP enzyme in Small intestine mucosa of Landrace were the maltose activity of duodenum, ileum mucosa were extremely significantly improved(P<0.01),the others were significantly decreased(P<0.05).4) It showed the SIg A of small mucosal morphology of Anqing Six White pigs and Landrace at the 33-day-old and 39-day-old that duodenum was the highest,followed by jejunum, ileum was the lowest, which reached a significant level.The SIg A of small mucosal morphology of Anqing Six White pigs were extremely significantly higher than that of Landrace at the 33-day-old and 39-day-old(P<0.01). Comparison of the same pig breeds, at the 39-day-old,the SIg A of small mucosal morphology of Anqing Six White pigs and Landrace were extremely significantly improved(P<0.01).5) According to the analysis of these 9 genes expression in small intestine between two pig breeds, there were significant or extremely significant difference in these 9 genes expression in small intestine between two pig breeds at the 33-day-old(P<0.05, 0.01);there were significant or extremely significant difference in FUT1, SLA-DQB, and TAP1 m RNA expression in small intestine between two pig breeds at the 33-day-old(P<0.05, 0.01); Comparison of the same Pig breeds, at the 39-day-old, the GLS, IGF-1, SLA-DQB m RNA expression levels in small intestine of Anqing Six White pigs were significantly or extremely significantly improved(P<0.05, 0.01),while the TAP1 and LYZ m RNA expression levels in small intestine of Anqing Six White pigs were significantly or extremely significantly decreased(P<0.05, 0.01); the GLS m RNA expression levels in small intestine of Landrace were extremely significantly or significantly improved(P<0.01, 0.05), the FUT1, TAP1, TLR4 and LYZ m RNA expression levels in small intestine of Landrace were extremely significantly or significantly decreased P<0.01, 0.05).6)At the 33-day-old and 39-day-old, There were positive correlation both between the GLS and EGFR m RNA expression levels in small intestine and Cor in the serum and between the IGF-1 and IGF-1R m RNA expression levels of small intestine and small intestine villus height of two pig breeds, some of small intestine reached a significant level(P< 0.05);there were significantly negative correlation between the FUT1, TLR4 m RNA expression levels in small intestine and T-AOC in the serum and the SIg A of small mucosal morphology, while there were positive correlation between the LYZ, SLA-DQB, TAP1 m RNA expression levels in small intestine and T-AOC in the serum and the SIg A of small mucosal morphology, some of small intestine reached a significant level(P< 0.05).From the above results, it is suggested that(1)Anqing Six White pigs and Landrace were weaned at the 28-day-old, there were differences of indicators of serum contents, intestine mucosal morphology, SIg A of small mucosal morphology at the 5d and the 10 d after weaning;(2)The GLS and EGFR gene play important roles in weaned piglets anti stress physiology reaction process, there were correlation between the IGF-1 and IGF-1R m RNA expression levels of small intestine and small intestine villus height, there were correlation between the decreased of FUT1, TLR4 m RNA expression levels in small intestine and the improved of LYZ, SLA-DQB, TAP1 m RNA expression levels in small intestine and the intestinal resistance of piglets during weaning stress stage.
