Helianthus tuberosus L.is an herbaceous plant from the Compositae family.Being aboundant with wide compatbility, low maintenace, easy planting and high fecundity, it is widely distribution in China. Helianthus tuberosus is a biological resource which is full of economic, ecological and social benefits and worth to further exploitation. The inulin which is rich in below-ground part of Helianthus tuberosus can be converted into fructose after hydrolysis, and the latter is a new type of edulcorator with high efficiency, safety and health protection. The leaf of Helianthus tuberosus has certain feeding value as it contains protein, carbohydrates and fiber.The study researched the production technology of high value-added products of Helianthus tuberosus on the purpose of industrial production.The production condition of extracting high fructose syrup from underground tubers and leaf protein concentration from fresh leaves are preliminary optimized. The main results are as follows:1.The inulin solution is extracted by hot water extraction.The effect of material liquid ratio, extraction temperature, time and times on inulin extraction rate is studied. The optimum extraction parameters determined through single factor and orthogonal test: material liquid is 1:25, extraction temperature is 90?, time is 30 min and extract one time. Under these conditions, the inulin extraction rate can reach 69.49%.2.Calcium hydroxide-phosphoric acid method is used to remove impurities such as protein and pectin in crude extract of inulin. The orthogonal experiment of removing impurity on the basis of single factor experiment show that the effect of calcium hydroxide dosage on protein removal is most significant.The best conditions for removing impurities: calcium hydroxide adjust pH value to 12, clarified temperature is 60?, clarified time is 20 min, phosphate adjust pH value to 7.5. Under these conditions, the protein removal rate of inulin crude extract is 78.90%, the average loss rate of total sugar is 7.42%.3.The fructose is obtained by inulinase one step hydrolysis from inulin extract.The effect of substrate concentration, enzyme amount, enzymolysis temperature and pH on the enzymolysis is studied. The best process conditions for enzymolysis:substrate concentration is 12-16%, enzyme dosage is 18?24 u·g-1, reaction pH is 4.5-5.0 and enzymolysis temperature 50 ?. Under these condition, the rate of enzymolysis can reach more than 95%.4.The dark crude extract liquid of high fructose syrup obtained after enzymolysis need decolorization and desalination. The active carbon and resin are used for further purification. In the study of active carbon, decoloring rate and total sugar loss rate are as the index, compares the effect of activated carbon dosage, decoloring time and temperature on decoloring effect. The best decoloring conditions determined:activated carbon dosage is 0.03 g·mL-1, decoloring time is 30 min and temperature is 80?. Under this condition, the decolorizing rate can reach more than 85%, and the total sugar loss is small.5.The syrup after active carbon treatment is refined by resin. The three indicator of adsorption rate, desalination rate and loss rate of total sugar are considered, and the resin with better effect is screened from eight kinds of resin through static adsorption method. Finally the 001 x 7 and D301-G are chosen.The coarse sugar liquid is treated by acidic cation resin type 001 x 7 firstly and then treated by alkaline anion resin type D301-G, the decoloring rate and the removal rate of Ca2+ and Mg2+ can reach more than 95%. The composition analysis of high fructose syrup shows that fructose content can reach 93.8%.6.The extraction process of leaf protein concentration from fresh Helianthus tuberosus leaf is studied. Compared different extraction agents for leaf protein extraction, and weigh three indicators of leaf protein yield, crude protein content and crude protein extraction rate, it is concluded that the extraction effect of sodium metabisulfite solution is best. On the basis of single factor experiment, the material liquid ratio, salt dosage, flocculation temperature and pH are dealt with orthogonal processing and the leaf protein concentratio extraction process was optimized. The best extraction conditions are concluded:material liquid is 1:7, salt dosage is 0.5%, flocculation temperature is 90? and pH value is 2.0. Under these conditions, the leaf protein yield is 12.77%,crude protein content is 48.96% and crude protein extraction rate is 27.36%. The amino acid analysis results of leaf protein show that the mino acid type of LPC is complete, total amino acid content is 42.07%, the ratio of essential amino acids to total amino acids (E/T) is 40.55%, the ratio of essential amino acids and non-essential amino(E/N) is 0.68.
